玉米HSP70基因家族的全基因組鑒定與分析

【摘要】:熱激蛋白70(HSP70)是生物體應(yīng)答各種脅迫反應(yīng)產(chǎn)生的一類蛋白,廣泛存在于生物體中,在幾乎所有的活細(xì)胞中都起著重要的作用。為全面解析玉米基因組HSP70基因信息,利用從Pfam數(shù)據(jù)庫(kù)中下載的隱馬氏模型文件PF00012在Gramene數(shù)據(jù)庫(kù)進(jìn)行基因搜索,并利用多種生物軟件或在線工具對(duì)所鑒定的基因進(jìn)行生物信息學(xué)分析。結(jié)果表明,試驗(yàn)共鑒定得到35個(gè)玉米HSP70基因,分別命名為Zm HSP70-1~Zm HSP70-35。這些基因不均衡地分布在玉米的10條染色體上,其中5號(hào)染色體上分布最多(6個(gè)),9號(hào)染色體上分布最少(1個(gè))。進(jìn)化樹分析顯示35個(gè)基因聚為6組,且同組基因具有相似的基因結(jié)構(gòu),其基序數(shù)量、類型和順序也是相似的。物種間進(jìn)化樹分析不同物種的86個(gè)基因,存在8對(duì)直系同源基因和17對(duì)旁系同源基因,直系同源基因中7對(duì)存在于玉米和水稻之間,僅1對(duì)存在于水稻和擬南芥之間,說(shuō)明起源于共同祖先基因的直系同源基因?qū)﹄m在單/雙子葉植物差異分離前就存在,但玉米與水稻間的進(jìn)化關(guān)系要比與擬南芥間更為親近。表達(dá)譜分析表明,大多數(shù)Zm HSP70基因?qū)俳M成型表達(dá),但表達(dá)模式和表達(dá)量不同。本研究結(jié)果為進(jìn)一步研究玉米HSP70家族基因的功能奠定了理論基礎(chǔ)。
[Abstract]:Heat shock protein 70 (HSP70) is a class of proteins produced by organism in response to various stress responses. It plays an important role in almost all living cells. In order to fully analyze the HSP70 gene information of maize genome. The hidden Markov model file PF00012, which was downloaded from Pfam database, was used for gene search in Gramene database. Bioinformatics analysis of the identified genes was carried out by using a variety of biological software or online tools. The results showed that 35 maize HSP70 genes were identified in the experiment. They were named Zm HSP70-1~Zm HSP70-35. These genes were distributed unevenly on 10 chromosomes of maize, of which chromosome 5 was the most distributed (6). The results of phylogenetic tree analysis showed that 35 genes were clustered into 6 groups and the same group had similar gene structure and the number of motifs. The phylogenetic tree of different species analyzed 86 genes of different species, and there were 8 pairs of lineal homologous genes and 17 pairs of collateral homologous genes. Seven pairs of lineal homologous genes existed between maize and rice, and only one pair existed between rice and Arabidopsis. The results indicated that the lineal homologous gene pairs originated from common ancestor genes existed before the differential separation of monocotyledonous plants, but the evolutionary relationship between maize and rice was closer than that between Arabidopsis and Arabidopsis. Most of the Zm HSP70 genes were constitutively expressed, but the expression patterns and expression levels were different. The results laid a theoretical foundation for further study on the function of HSP70 family genes in maize.
