喹高利特對(duì)大鼠乳腺乳脂合成相關(guān)基因調(diào)控作用的研究

【摘要】:催乳素是在泌乳啟動(dòng)和維持中起重要作用的激素。催乳素可以促進(jìn)乳腺導(dǎo)管延伸、乳汁生成,促進(jìn)乳成分的合成,尤其影響乳脂的合成。為了研究催乳素對(duì)乳脂合成的調(diào)控機(jī)制,本研究使用多巴胺(D2)受體激動(dòng)劑喹高利特抑制泌乳大鼠催乳素的釋放,采用數(shù)字基因表達(dá)譜分析比對(duì)喹高利特處理組(Quinagolide)與對(duì)照組(Control)大鼠泌乳乳腺乳脂合成基因的差異表達(dá)情況。為了驗(yàn)證測(cè)序結(jié)果的準(zhǔn)確性,本研究選取了10個(gè)基因,使用熒光定量PCR技術(shù)測(cè)定了其表達(dá)情況。本試驗(yàn)使用轉(zhuǎn)錄組測(cè)序的方法構(gòu)建了兩組(Quinagolide、Control)大鼠乳腺的總RNA文庫(kù),平均產(chǎn)生14,458,067條clean reads。兩個(gè)文庫(kù)分別得到12843和13639個(gè)基因。以|log2Ratio(Quinagolide/Control)|≥2,FDR≤0.001為基因顯著差異標(biāo)準(zhǔn),分析兩組乳腺組織基因差異表達(dá)情況。對(duì)照組與喹高利特處理組比對(duì),顯著差異表達(dá)的基因共有2823個(gè),其中下調(diào)基因262個(gè),上調(diào)基因2561個(gè),分別占到差異表達(dá)基因總數(shù)的3.9%和42.3%。轉(zhuǎn)錄水平的研究結(jié)果表明:(1)喹高利特抑制催乳素主要影響乳腺上皮細(xì)胞分化、生長(zhǎng),進(jìn)而影響乳產(chǎn)量。(2)催乳素抑制直接調(diào)節(jié)脂肪酸結(jié)合蛋白(FABP3)、乙酰輔酶A羧化酶(ACC)、脂肪酸脫氫酶(FADS1)、胰島素誘導(dǎo)基因1(INSIG1)的轉(zhuǎn)錄水平。
[Abstract]:Prolactin is a hormone that plays an important role in the initiation and maintenance of lactation. Prolactin can promote breast duct extension, milk production and the synthesis of milk components. In order to study the regulation mechanism of prolactin on the synthesis of milk fat, we used the dopamine D _ 2) receptor agonist quinolide to inhibit the prolactin release in lactating rats. Quinagolide (Quinagolide) and control group (control) were compared by digital gene expression profile analysis. Differential expression of milk fat synthesis gene in rat lactation mammary gland in order to verify the accuracy of sequencing results. In this study, 10 genes were selected and their expression was determined by fluorescence quantitative PCR. Two groups of Quinagolide were constructed by transcriptome sequencing.
