羥基磷灰石表面構(gòu)建壓電陶瓷涂層的生物相容性研究

【摘要】:目的:評(píng)價(jià)羥基磷灰石表面構(gòu)建的壓電陶瓷涂層的生物相容性。方法:按照ISO/10993及GB/T16886系列標(biāo)準(zhǔn),對(duì)羥基磷灰石表面構(gòu)建的壓電陶瓷涂層進(jìn)行檢測(cè)。1.溶血試驗(yàn):在稀釋抗凝兔血中分別加入羥基磷灰石表面構(gòu)建的壓電陶瓷涂層浸提液、生理鹽水與蒸餾水,檢測(cè)溶血率;2.短期全身毒性試驗(yàn)(經(jīng)口途徑):對(duì)大鼠分別灌胃給予羥基磷灰石表面構(gòu)建的壓電陶瓷涂層浸提液與生理鹽水,觀察動(dòng)物每周的體重變化,食物利用率;3.熱原試驗(yàn):自新西蘭兔耳緣靜脈分別注射羥基磷灰石表面構(gòu)建的壓電陶瓷涂層浸提液與生理鹽水,觀察動(dòng)物體溫變化。4.致敏試驗(yàn):試驗(yàn)組以羥基磷灰石表面構(gòu)建的壓電陶瓷涂層浸提液與弗氏完全佐劑為供試液,對(duì)照組以生理鹽水溶液與弗氏完全佐劑為供試液。用最大劑量法進(jìn)行致敏試驗(yàn),觀察動(dòng)物的皮膚情況。結(jié)果:1.羥基磷灰石表面構(gòu)建的壓電陶瓷涂層浸提液的溶血率為0.86%;2.灌胃一周后,試驗(yàn)組與對(duì)照組大鼠體重增長(zhǎng)量差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05),食物利用率分別為16.5%,16.9%,灌胃兩周后,試驗(yàn)組與對(duì)照組大鼠體重增長(zhǎng)量差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05),食物利用率分別為16.71%,17.1%;3.注射羥基磷灰石表面構(gòu)建的壓電陶瓷涂層浸提液和生理鹽水后,試驗(yàn)組兔體溫升高0.30℃,試驗(yàn)組兔體溫升高總和為0.60℃;4.去除貼敷物24,48,72小時(shí)后,試驗(yàn)組與對(duì)照組豚鼠的皮膚評(píng)分均小于1。結(jié)論:羥基磷灰石表面構(gòu)建的壓電陶瓷涂層無(wú)溶血作用、無(wú)毒性、不致熱、無(wú)致敏作用,具有良好的生物相容性。
[Abstract]:Objective: to evaluate the biocompatibility of piezoelectric ceramic coating on hydroxyapatite. Methods: according to ISO/10993 and GB/T16886 series standards, the piezoelectric ceramic coating on hydroxyapatite surface was tested. Hemolysis test: dilute anticoagulant rabbit blood by adding hydroxy apatite on the surface of piezoelectric ceramic coating extract, physiological saline and distilled water, to detect the hemolysis rate of 2. 5%. Short-term systemic toxicity test (via oral approach: rats were given hydroxy apatite surface piezoelectric ceramic coating extract and physiological saline respectively, to observe the weekly body weight changes, food utilization rate of 3. 5%. Pyrogen test: piezoelectric ceramic coating extract and physiological saline were injected into the auricular vein of New Zealand rabbit respectively. The changes of body temperature of animals were observed. Sensitization test: the piezoelectric ceramic coating extractant and Freund's complete adjuvant on hydroxyapatite were used as the test solution in the test group, and the physiological saline solution and Freund's complete adjuvant in the control group were used as the test solution. The maximum dose method was used to sensitize animals to observe the skin condition. The result is 1: 1. The hemolysis rate of the piezoelectric ceramic coating solution constructed on hydroxyapatite is 0.86% and 2% respectively. After a week of gavage, there was no significant difference in body weight growth between the experimental group and the control group (P 0.05), but the food utilization rate was 16. 5% and 16. 9% respectively. After two weeks of gavage, there was no significant difference in body weight growth between the experimental group and the control group (P 0. 05), and the food utilization rate was 16. 71% and 17. 1% respectively.
