超高效液相色譜-串聯(lián)質譜法檢測微氧生物脫氮菌群酰基高絲氨酸內酯信號分子

【摘要】:為揭示處理低碳氮比廢水的微氧活性污泥系統(tǒng)的生物脫氮機制,了解脫氮功能菌群的群體生長和代謝規(guī)律,建立了超高效液相色譜-串聯(lián)質譜同時定量檢測介導革蘭氏陰性(G!)細菌群體感應信號分子;呓z氨酸內酯(AHLs)的方法。取自升流式微氧活性污泥反應器的泥水混合物,使用乙酸乙酯液液萃取,旋轉蒸干后以甲醇定容,經(jīng)C18色譜柱分離。以5 mmol/L乙酸銨(含0.1%甲酸)和甲醇為流動相進行梯度洗脫,采用多反應離子監(jiān)測模式,使用配有電噴霧離子源的三重四極桿質譜進行檢測。對9種AHLs的檢測結果表明,在0.5~100μg/L范圍內呈現(xiàn)良好的線性關系,檢出限為0.01~0.5μg/L,回收率為62.5%~118.1%,相對標準偏差為2.9%~12.1%,分析時間為6.5 min。本方法具有快速、準確和精密等特點,可及時反映活性污泥功能菌群的生長狀態(tài)和代謝活性,對了解生物脫氮系統(tǒng)的生物學機制和廢水生物處理系統(tǒng)的運行調控具有重要意義。
