乳酶生菌種脂肪酸組分的GC-MS檢測(cè)分析

【摘要】:目的:建立氣相色譜-質(zhì)譜聯(lián)用法測(cè)定乳酶生菌種的脂肪酸成分,并對(duì)各試樣菌株間的同源性進(jìn)行分析。方法:應(yīng)用14%三氟化硼-甲醇法對(duì)菌株進(jìn)行脂肪酸甲酯化,采用HP-5MS毛細(xì)管色譜柱(30 m×0.250 mm×0.25μm),載氣為氦氣,程序升溫(120℃,5 min;6℃·min~(-1)升至240℃,10 min;10℃·min~(-1)至260℃,2 min),進(jìn)樣口溫度250℃,選擇EI離子源,溶劑延遲3 min,質(zhì)譜掃描m/z 35~450。結(jié)果:腸球菌屬的菌株均具有十四碳烷酸、十六碳烷酸、十六碳烯酸、十八碳烷酸、十八碳烯酸及十九碳烷酸特征峰;批號(hào)為20100922和20101001的乳酶生片菌株與海氏腸球菌CGMCC1.595之間,6家生產(chǎn)企業(yè)的其他乳酶生片菌株與糞腸球菌140623和屎腸球菌CGMCC1.131之間,具有相似的氣相色譜圖及相近的脂肪酸組分比值。結(jié)論:本方法可用于乳酶生菌種的快速鑒定及同源性分析。
