HPLC同時測定雞血藤中芒柄花素和美迪紫檀素等6個黃酮類成分

【摘要】:目的:建立HPLC同時測定雞血藤中原兒茶酸、兒茶素、表兒茶素、甘草素、芒柄花素、美迪紫檀素含量的方法,為雞血藤藥材質(zhì)量標準改進提供參考。方法:采用ZORBAX SB-C18色譜柱(4.6 mm×150 mm,5μm),以乙腈(A)-0.2%磷酸(B)為流動相進行梯度洗脫,流速1 m L·min~(-1),檢測波長為260 nm(檢測原兒茶酸、芒柄花素)和280 nm(檢測兒茶素、表兒茶素、甘草素、美迪紫檀素),柱溫30℃。結(jié)果:原兒茶酸、兒茶素、表兒茶素、甘草素、芒柄花素、美迪紫檀素的質(zhì)量濃度分別在2.286~228.6μg·m L~(-1)(r=0.999 9)、2.174~217.4μg·m L~(-1)(r=0.999 9)、2.79~279μg·m L~(-1)(r=0.999 9)、0.424~42.4μg·m L~(-1)(r=0.999 9)、2.426~242.6μg·m L~(-1)(r=0.999 9)、0.339~33.92μg·m L~(-1)(r=0.999 8)的范圍內(nèi)線性關(guān)系良好,加樣回收率分別為99.8%(RSD=1.0%)、99.8%(RSD=2.5%)、101.2%(RSD=2.3%)、100.8%(RSD=1.8%)、100.9%(RSD=2.3%)、100.6%(RSD=2.4%)。11批藥材中原兒茶酸、兒茶素、表兒茶素、甘草素、芒柄花素、美迪紫檀素含量范圍分別為0.250~0.898、1.447~5.662、1.759~11.925、0.035~0.134、0.218~0.535、0.065~0.585 mg·g~(-1)。結(jié)論:該方法可用于同時測定雞血藤中6個黃酮類成分的含量,為該藥材的質(zhì)量標準改進提供參考。
