有機雙光子線粒體內(nèi)活性小分子熒光探針研究進展

【摘要】:線粒體在細胞的能量代謝中發(fā)揮著關(guān)鍵作用,其內(nèi)部環(huán)境的微小變化會影響細胞的正常生命活動。同時線粒體內(nèi)許多活性小分子在細胞的許多生理過程中也起著關(guān)鍵作用。因此,可視化監(jiān)測線粒體自身及內(nèi)部微環(huán)境的變化對于生命現(xiàn)象的研究和疾病的診斷與治療具有非常重要的意義。熒光分析法因具有操作簡便、靈敏度高、選擇性好、實時檢測以及損傷小等優(yōu)點而得到了廣泛的研究和應用。雙光子熒光探針技術(shù)相對于單光子熒光技術(shù)具有長波吸收短波發(fā)射、高度的三維空間選擇性、大的穿透深度、避免熒光漂白和光致毒以及降低組織自發(fā)熒光干擾等特點,在生命科學領域具有廣闊的應用前景。該文介紹了有機雙光子吸收基本原理以及有機雙光子線粒體內(nèi)活性小分子熒光探針的研究現(xiàn)狀,同時對有機線粒體內(nèi)活性小分子熒光探針今后的發(fā)展趨勢進行了展望。
[Abstract]:Mitochondria play a key role in the energy metabolism of cells. At the same time, many active small molecules in mitochondria also play a key role in many physiological processes of cells. Therefore, it is very important for the study of life phenomena and the diagnosis and treatment of diseases to monitor the changes of mitochondria itself and internal microenvironment. Fluorescence analysis has been widely studied and applied because of its advantages of simple operation, high sensitivity, good selectivity, real-time detection and small damage. Compared with single photon fluorescence technique, two-photon fluorescence probe has the advantages of long wave absorption shortwave emission, high three-dimensional selectivity, large penetration depth, avoiding fluorescent bleaching and phototoxicity, and reducing tissue autofluorescence interference. It has a broad application prospect in the field of life science. In this paper, the basic principle of organic two-photon absorption and the research status of active small molecule fluorescence probes in organic two-photon mitochondria are introduced. The development trend of active small molecule fluorescence probes in organic mitochondria is also prospected.
