親水作用色譜-質(zhì)譜法測(cè)定麻黃根多糖單糖的組成

【摘要】:目的:建立一種親水相互作用色譜-質(zhì)譜(HILIC-UPLC-MS/MS)檢測(cè)分析方法,用于直接檢測(cè)麻黃根多糖的單糖組成。方法:采用ACQUITY UPLC BEH Amide色譜柱(2.1 mm×100 mm,1.7μm),流動(dòng)相A為乙腈-水(95∶5),B為乙腈-水(5∶95),柱溫60℃,梯度洗脫(0~10 min,92%~70%A;10~10.1 min,70%~92%A;10.1~15 min,92%A),進(jìn)樣量2μL,流速0.2 m L·min~(-1);質(zhì)譜采用電噴霧離子源,負(fù)離子模式下多反應(yīng)離子檢測(cè)。Turbo V離子源參數(shù)如下:毛細(xì)管電壓-4.5 k V,源溫度550℃;氣簾氣和碰撞氣壓力分別設(shè)為30 psi和10 psi;霧化氣和氣化氣壓力均為55 psi,碰撞電池入口電位和出口電位均為-10 V。結(jié)果:該方法在12 min內(nèi)實(shí)現(xiàn)8種未衍生單糖成分的完全基線分離,精密度、穩(wěn)定性和重復(fù)性的RSD均2.1%,各成分的回收率在99.3%~102.5%,RSD在1.5%~2.9%,應(yīng)用上述建立的方法分別對(duì)5批麻黃根多糖進(jìn)行單糖組成分析,結(jié)果顯示,麻黃根中檢測(cè)到5種單糖分別為L(zhǎng)-鼠類糖,L-阿拉伯糖,D-甘露糖,D-半乳糖,D-半乳糖醛酸。結(jié)論:HILICUPLC-MS/MS方法靈敏度高、重復(fù)性好、快速、準(zhǔn)確、樣品前處理簡(jiǎn)單、無(wú)需衍生化,可用于植物多糖中單糖及寡糖的快速檢測(cè)。
