吳茱萸UPLC指紋圖譜的建立及質(zhì)量評價

【摘要】:目的:建立吳茱萸超高效液相色譜(UPLC)指紋圖譜分析方法,對不同品種、不同產(chǎn)地吳茱萸藥材的質(zhì)量進(jìn)行較全面的評價。方法:采用ACQUITY UPLC BEH C18色譜柱(2.1 mm×100 mm,1.7μm),以乙腈-0.1%甲酸水為流動相進(jìn)行梯度洗脫,檢測波長300,215 nm,流速0.4 m L·min~(-1),柱溫35℃。采用相似度評價、聚類分析及偏最小二乘法判別分析對37批樣品的指紋圖譜進(jìn)行分析。結(jié)果:建立了吳茱萸藥材的UPLC指紋圖譜及共有模式,標(biāo)定了34個共有峰,采用對照品比對,指認(rèn)出17個共有峰。37批樣品的相似度在0.703~0.973。不同來源的吳茱萸藥材指紋圖譜輪廓基本一致,化學(xué)成分的相對含量在較大范圍內(nèi)波動。聚類分析和偏最小二乘判別分析結(jié)果表明,化學(xué)成分的相對含量與藥材品種、產(chǎn)地沒有明顯的相關(guān)性,而與藥材的千粒重在一定程度上相關(guān);吳茱萸次堿、吳茱萸堿、新綠原酸、綠原酸、吳茱萸卡品堿、檸檬苦素是差異較大的6個成分。結(jié)論:該方法簡便、快速,可有效、全面地用于吳茱萸藥材的質(zhì)量評價;控制吳茱萸藥材質(zhì)量的關(guān)鍵是把握"近成熟"這一采收時間環(huán)節(jié)。
