席夫堿型熒光探針的合成及其光譜性質(zhì)的研究

【摘要】:隨著人類各金屬用品的使用以及環(huán)境污染的加劇,使生物體內(nèi)金屬離子的含量都有所增加,并可能造成生物體各系統(tǒng)的紊亂,甚至?xí)鸶鞣N疾病的發(fā)生,嚴(yán)重影響生命體的健康。為了檢測金屬離子的存在以及含量,需要一個(gè)高效并且能夠快速檢測金屬離子的方法。熒光探針因有良好的選擇性、識別性、快速檢測性及在環(huán)境與生物體中的可應(yīng)用性,而受到廣大研究者的關(guān)注。席夫堿類探針合成簡單、配位能力強(qiáng),是組成探針理想的候選結(jié)構(gòu)。本文合成了6種不同的席夫堿型熒光探針分子,通過HRMS,IR,1HNMR以及13CNMR對探針化合物的結(jié)構(gòu)進(jìn)行表征,并對其光譜性質(zhì)進(jìn)行了研究。具體內(nèi)容如下:1.對探針的結(jié)構(gòu)、影響因素以及機(jī)理等方面進(jìn)行了綜述,重點(diǎn)對近年報(bào)道的席夫堿型熒光探針類做了總結(jié)。2.以羅丹明B為熒光團(tuán),設(shè)計(jì)了4種席夫堿型熒光探針R1、R2、R4、R5,主要測試了包括離子選擇性、競爭性、濃度滴定曲線、Job曲線、pH及時(shí)間對探針熒光強(qiáng)度的影響等項(xiàng)目,并且討論了取代基對探針熒光強(qiáng)度的影響。通過紫外吸收和熒光光譜的測定表明,4種產(chǎn)物可以作為Fe~(3+)探針。與探針R3相比,探針R1的熒光強(qiáng)度減弱,探針R2的熒光強(qiáng)度增加。探針R4、R5對Fe~(3+)的選擇識別能力很高,對其他離子無響應(yīng)。3.以噻唑類衍生物為電子供體,合成了席夫堿型探針化合物R6、R7。通過研究發(fā)現(xiàn),Cd~(2+)的加入會使探針R6熒光迅速增強(qiáng),實(shí)現(xiàn)高選擇性的識別,而且探針R6與Cd~(2+)體系可以組成新的熒光探針RC實(shí)現(xiàn)接力識別。探針RC可以通過熒光猝滅的方式高效識別PO_4~(3-),其他陰離子對其檢測沒有干擾。當(dāng)PO_4~(3-)在一定濃度范圍內(nèi)時(shí),探針RC的熒光強(qiáng)度與PO_4~(3-)濃度具有一定的線性關(guān)系,可以定量檢測PO_4~(3-)。而探針R7則對Fe~(3+)具有一定的的選擇性。
[Abstract]:With the use of human metal supplies and the intensification of environmental pollution, the content of metal ions in organisms has increased, which may lead to the disorder of biological systems and even the occurrence of various diseases. Seriously affect the health of life. In order to detect the presence and content of metal ions, an efficient and rapid method is needed to detect metal ions. Fluorescent probes have attracted much attention due to their good selectivity, recognition, rapid detection and applicability in environment and organisms. Schiff base probe is an ideal candidate structure for its simple synthesis and strong coordination ability. In this paper, six kinds of Schiff base fluorescent probe molecules were synthesized and characterized by HRMS,IR,1HNMR and 13CNMR, and their spectral properties were studied. The specific contents are as follows: 1. The structure, influencing factors and mechanism of the probes are reviewed, and the Schiff base fluorescent probes reported in recent years are emphatically summarized. 2. Using Rhodamine B as the fluorescence group, four kinds of Schiff base fluorescence probes R1OR2OR4OR5 have been designed. The main items include ion selectivity, competition, concentration titration curve, Job curve, pH and the influence of time on the fluorescence intensity of the probe. The influence of substituents on the fluorescence intensity of the probe was also discussed. The UV absorption and fluorescence spectra showed that the four products could be used as Fe~ (3) probes. Compared with probe R3, the fluorescence intensity of probe R1 decreased and the fluorescence intensity of probe R2 increased. The probe R4N R5 has a high ability to select and recognize Fe~ (3), and has no response to other ions. 3. The Schiff base probe compound R 6 O R 7 was synthesized by using thiazole derivatives as electron donor. It is found that the fluorescence of probe R6 can be enhanced rapidly with the addition of Cd~ (2), and the system of probe R6 and Cd~ (2) can form a new fluorescent probe RC to realize relay recognition. The probe RC can efficiently recognize PO_4~ (3-) by fluorescence quenching, and other anions do not interfere with the detection of PO_4~ (3-). When PO_4~ (3-) is in a certain concentration range, the fluorescence intensity of the probe RC has a certain linear relationship with the concentration of PO_4~ (3-). It can be used to quantitatively detect PO_4~ (3-). The probe R7 has a certain selectivity to Fe~ (3).
