HPLC-ELSD法測定三種植物基原金線蓮的金線蓮苷含量

【摘要】:建立HPLC-ELSD法,測定三種植物基原金線蓮中金線蓮苷含量。采用phenomenex NH2色譜柱(250 mm×4.6 mm,5μm),以乙腈-水(92∶8,V/V)為流動相進行等度洗脫,流速為0.8 m L/min,柱溫室溫,ELSD霧化室溫度為70℃,氮氣流速為1.5 L/min。實驗結(jié)果表明,金線蓮苷進樣量在76.02~2432.64μg/m L范圍內(nèi)線性良好(r=0.9991);精密度、穩(wěn)定性、重復(fù)性實驗的RSD均≤3%;平均加樣回收率為97.55%,RSD為1.50%(n=6)。33批次金線蓮的金線蓮苷含量在6.37%~22.66%之間,平均含量為15.00%,存在較大的差異(RSD=26.29%)。該方法快速、簡便,結(jié)果準(zhǔn)確、可靠,適用于金線蓮中金線蓮苷的定量分析。
