高效液相色譜法測(cè)定大豆中六種異黃酮含量方法的改進(jìn)

【摘要】:為探索大豆異黃酮乙醇提取液的高效液相色譜檢測(cè)方法,本研究按照改進(jìn)后的色譜條件對(duì)分析方法學(xué)進(jìn)行了評(píng)價(jià)。確定的色譜條件如下,流動(dòng)相為0.1%乙酸-水溶液和0.1%乙酸-乙腈溶液,梯度洗脫;柱溫30℃;檢測(cè)波長(zhǎng)260 nm;流速1.0 m L/min。六種大豆異黃酮線性范圍良好,決定系數(shù)R20.9993,回收率范圍99.62%~114.00%,相對(duì)標(biāo)準(zhǔn)偏差RSD范圍0.96%~7.99%。本試驗(yàn)改進(jìn)的檢測(cè)方法線性關(guān)系、精密度、穩(wěn)定性、準(zhǔn)確度、靈敏度均較好,滿足分析方法學(xué)評(píng)價(jià)的要求及大豆中六種異黃酮單體檢測(cè)的要求,為改進(jìn)大豆中異黃酮的檢測(cè)方法提供了參考。
