苯并咪唑硫醚銀配合物的合成及生物性質(zhì)研究

【摘要】:由于金屬銀及其配合物具有良好的殺菌抗菌性質(zhì),近些年來(lái)引起了人們的興趣與廣泛關(guān)注。銀離子的配位數(shù)從2到6,從而使它的幾何構(gòu)型既有平面線(xiàn)性構(gòu)型,也有八面體構(gòu)型。作為典型的雜環(huán)配體,苯并咪唑配體展現(xiàn)出了優(yōu)良的光物理化學(xué)性質(zhì)及良好的抗病毒性質(zhì)。本論文選擇苯并咪唑硫醚為配體,與不飽和烯酸銀合成結(jié)構(gòu)新穎的銀配合物,并研究配合物結(jié)構(gòu)與性質(zhì)的關(guān)系。本文設(shè)計(jì)合成了四個(gè)苯并咪唑硫醚配體S1-S~4,與不同的烯酸銀鹽反應(yīng)(甲基丙烯酸銀,肉桂酸銀,鄰香豆酸銀,對(duì)香豆酸銀,水楊酸銀,巴豆酸銀),制備出七個(gè)不同的銀配合物。用現(xiàn)代分析方法對(duì)其結(jié)構(gòu)進(jìn)行表征,七個(gè)不同的銀配位化合物組成如下:[Ag(S~2)(p-coumarate)]·2CH_3OH(1)[Ag(S~3)(methacrylate)](2)[Ag(S~3)(cinnamate)](3)[Ag(S~3)_2](o-coumarate)·CH_3CN·2C_2H_5OH(4)[Ag(S~3)(crotonate)](5)[Ag(S~4)(crotonate)](6){[Ag_4(μ-4,4′-bipy)3(μ-salicylate)2(salicylate)H_2O]2?2C_2H_5OH}_n(7)X-射線(xiàn)單晶衍射表明配合物1、2、3、5、6結(jié)構(gòu)相似,它們都是單核三配位的平面三角形結(jié)構(gòu),配合物4是單核四配位的扭曲四面體結(jié)構(gòu),配合物7是一個(gè)無(wú)機(jī)聚合物,并且是三核銀簇化合物。尤其是在無(wú)機(jī)聚合物7中,每個(gè)聚合單位由中心對(duì)稱(chēng)的兩部分組成,每個(gè)部分包含四個(gè)一價(jià)銀離子,三個(gè)聯(lián)吡啶配體,三個(gè)水楊酸陰離子和一個(gè)水分子,4,4′-聯(lián)吡啶作為橋聯(lián)配體與其他單元相連接,形成了無(wú)限延伸的一維鏈狀聚合物。水楊酸陰離子有兩種配位方式:一種是作為橋聯(lián)配體與銀(I)進(jìn)行雙齒配位,連接Ag1,Ag3,Ag_4;另外一種是與Ag2單齒配位,提供配位點(diǎn)。以Ag1為中心,周?chē)鷰讉(gè)原子形成了扭曲的四方錐體,四方錐的變形參數(shù)是τ5,用于形容五配位的金屬體系變形系數(shù)。以Ag1為中心的四方錐參數(shù)τ5的數(shù)值是0.11;Ag2是三配位的,與周?chē)訕?gòu)成了一個(gè)平面三角形;Ag3和Ag_4都是四配位的,與周?chē)牡踉有纬伤拿骟w幾何構(gòu)型。應(yīng)用紫外滴定,熒光滴定,粘度測(cè)試等實(shí)驗(yàn)方法,研究了配體、配合物與DNA之間的相互作用模式,測(cè)定了減色率,線(xiàn)性擬合常數(shù)Kb,熒光淬滅常數(shù)Ksv,實(shí)驗(yàn)數(shù)據(jù)表明,配體及配合物與DNA的作用模式是插入模式,配合物與DNA的結(jié)合能力要強(qiáng)于配體。使用Fetion法及NBT光照法研究了銀配合物抗羥基自由基和抗超氧自由基的活性,用IC50值表征配合物清除自由基能力的大小。研究結(jié)果表明配合物1、4、5、7的IC50值比標(biāo)準(zhǔn)的VC和甘露醇低兩個(gè)數(shù)量級(jí),說(shuō)明配合物1、4、5、7有更優(yōu)異的清除羥基自由基的能力;而配合物1、7的IC50值比SOD標(biāo)準(zhǔn)模擬復(fù)合物小,說(shuō)明配合物1、7可以作為SOD模擬酶清除超氧陰離子。
[Abstract]:Silver and its complexes have attracted much attention in recent years due to their good bactericidal and antibacterial properties. The coordination number of silver ions ranges from 2 to 6, which results in both plane linear configuration and octahedral configuration. As typical heterocyclic ligands, benzimidazole ligands exhibit excellent photophysicochemical and antiviral properties. In this paper, benzimidazole sulfides were selected as ligands to synthesize novel silver complexes with silver unsaturated enoate, and the relationship between the structure and properties of the complexes was studied. In this paper, four benzimidazolyl thioether ligands S _ (1-S) _ (4) were designed and synthesized.
