銅綠假單胞菌SJTD-1的磷酸化蛋白質組學

【摘要】:制備了銅綠假單胞菌SJTD-1菌株的蛋白質組酶解產物,并利用二氧化鈦磁珠從酶解產物中富集磷酸化肽.首次利用納升液相色譜-質譜技術鑒定磷酸化肽的磷酸化位點,最終共鑒定獲得13條磷酸化肽,分別來自12個磷酸化蛋白,其中有7個磷酸化蛋白在假單胞菌中是首次發(fā)現(xiàn).分析鑒定到的磷酸化蛋白,發(fā)現(xiàn)大多與DNA復制、物質轉運、能量代謝等密切相關.這是首次對銅綠假單胞菌SJTD-1菌株進行磷酸化蛋白質組學分析,為SJTD-1在DNA復制、物質和能量代謝以及細胞膜轉運等方面的研究奠定基礎.
[Abstract]:The proteomic hydrolysis products of Pseudomonas aeruginosa (SJTD-1) strain were prepared and the phosphorylated peptides were enriched by titanium dioxide magnetic beads. The phosphorylation sites of phosphorylated peptides were identified by nanolith liquid chromatography-mass spectrometry for the first time. A total of 13 phosphorylated peptides were identified, each from 12 phosphorylated proteins, of which 7 phosphorylated proteins were first found in Pseudomonas. Most of the phosphorylated proteins identified were closely related to DNA replication, substance transport and energy metabolism. This is the first time that the phosphorylation proteomics analysis of Pseudomonas aeruginosa SJTD-1 strain is carried out, which lays a foundation for the study of SJTD-1 in DNA replication, substance and energy metabolism and cell membrane transport.
