制備色譜法分離純化高良姜黃酮中高良姜素和山柰素

【摘要】:建立了制備型高效液相色譜(Prep-HPLC)分離高良姜黃酮中高良姜素和山柰素的方法。高良姜黃酮經(jīng)HPD-600樹脂吸附洗脫純化后,采用Prep-HPLC分離高良姜黃酮中高良姜素和山柰素。制備色譜條件:流動相為甲醇-0.6%(v/v)乙酸水溶液(58∶42,v/v),柱溫為常溫,流速為7.0 mL/min,檢測波長為360 nm,進樣量為700μL,上樣質(zhì)量濃度為10.0 g/L。分離的單體由質(zhì)譜和核磁共振氫譜、碳譜鑒定確證為高良姜素和山柰素,HPLC外標法定量,純度分別為99.5%和99.7%。該方法分離效果好、高效、低毒,可用于高良姜中高良姜素和山柰素的分離制備。
[Abstract]:A method for the separation of galangin and kaempferol from flavonoids of alpinia officinalis by Prep-HPLC was established. The flavonoids of alpinia officinalis were eluted and purified by HPD-600 resin, and Prep-HPLC was used to separate alpinol and kaempferol from the flavonoids of alpinia officinalis. The chromatographic conditions were as follows: the mobile phase was methanol-0.6% (v / v) acetic acid solution (58: 42 v / v), the column temperature was normal temperature, the flow rate was 7.0 mL/min, the detection wavelength was 360 nm, the sample volume was 700 渭 L, and the sample concentration was 10.0 g / L. The isolated monomers were identified by mass spectrometry and nuclear magnetic resonance (NMR) and identified as gingerin and kaempferol by HPLC. The purity was 99.5% and 99.7%, respectively. The method has the advantages of good separation, high efficiency and low toxicity. It can be used for the separation and preparation of galangin and kaempferol from alpinia officinalis.
