10個采收期青錢柳HPLC指紋圖譜建立及4種成分測定

【摘要】:目的建立10個采收期(3月末、5月初、7月初、8月末、9月初、9月末、10月初、10月末、11月初、11月中)青錢柳Cyclocarya paliurus(Batal)Iljinsk的HPLC指紋圖譜,并測定4種成分含有量。方法青錢柳80%甲醇提取液的分析采用Agilent C18色譜柱(5μm,250 mm×4.6 mm);乙腈-0.1%磷酸為流動相,梯度洗脫;檢測波長365nm(指紋圖譜)和254 nm(含有量測定);體積流量1.0 m L/min;柱溫35℃。再對指紋圖譜中的共有峰進行主成分分析。結(jié)果 10批樣品指紋圖譜中有15個共有峰,相似度0.920~0.981,4個主成分累計貢獻率達92.82%,峰1、2(綠原酸)、12、14的權(quán)重值較大。綠原酸、異槲皮苷、槲皮素、山柰酚分別在0.008 492~0.566 1、0.004 370~0.291 3、0.000 551 5~0.036 77、0.001 287~0.085 77 mg/m L范圍內(nèi)線性關(guān)系良好,平均加樣回收率分別為98.2%、97.5%、96.3%、97.0%,RSD分別為1.35%、1.14%、2.21%、1.74%。各采收期樣品中4種成分含有量差異明顯,其中綠原酸與異槲皮苷在7月初最高,而槲皮素和山柰酚分別在8月末和9月初最高。結(jié)論該方法可用于青錢柳的質(zhì)量控制,并為篩選其最佳采收期提供依據(jù)。
