外源β-葡萄糖苷酶處理結(jié)合異煙酸-吡唑啉酮分光光度法測定橡膠籽中氰化物含量

【摘要】:研究經(jīng)不同方法脫毒處理后的橡膠籽分別采用異煙酸-吡唑啉酮分光光度法和加入外源β-葡萄糖苷酶的異煙酸-吡唑啉酮分光光度法檢測其氰化物含量。結(jié)果表明:采用異煙酸-吡唑啉酮分光光度法檢測出的氰化物含量較低或不存在,而加入外源β-葡萄糖苷酶的異煙酸-吡唑啉酮分光光度法檢測出的氰化物含量明顯高于單獨(dú)采用異煙酸-吡唑啉酮分光光度法的檢出值。再對加入外源β-葡萄糖苷酶的異煙酸-吡唑啉酮分光光度法的應(yīng)用條件進(jìn)行優(yōu)化后得到最優(yōu)條件為浸泡時間3.35 h、浸泡溫度45.06℃、加酶量24.11 U/100 mL、浸泡pH 5.56。以上條件下,預(yù)測的氰化物含量為28.79μg/g,加標(biāo)回收率為85.02%~92.04%,測定結(jié)果準(zhǔn)確可靠。
[Abstract]:Isonicotinic acid-pyrazolone spectrophotometry and isonicotinic acid-pyrazolone spectrophotometry were used to determine the cyanide content of rubber seeds treated with different detoxification methods. The results showed that the cyanide content detected by isonicotinic acid-pyrazolone spectrophotometry was low or non-existent. The content of cyanide detected by isonicotinic acid-pyrazolone spectrophotometry was significantly higher than that by isonicotinic acid-pyrazolone spectrophotometry.
