基于化學(xué)指紋圖譜和多指標測定的柿葉藥材質(zhì)量研究

【摘要】:目的建立柿Diospyros kaki葉的HPLC指紋圖譜,并進行多成分測定,為柿葉及其相關(guān)產(chǎn)品的質(zhì)量控制提供科學(xué)依據(jù)。方法采用HPLC法,Diamonsil C18(250 mm×4.6 mm,5μm)色譜柱;以乙腈-0.2%甲酸水為流動相梯度洗脫;體積流量1.0 m L/min;柱溫35℃;檢測波長360 nm;進樣量10μL;對22批柿葉藥材進行指紋圖譜研究,用中藥色譜指紋圖譜相似度評價系統(tǒng)(2004A版)進行評價,系統(tǒng)聚類分析和PCA分析將22批柿葉藥材分為兩類,同時對金絲桃苷、異槲皮苷、槲皮素和山柰素成分測定方法進行方法學(xué)考察。結(jié)果建立了柿葉的指紋圖譜,共標定了12個色譜峰為柿葉藥材指紋圖譜的特征峰,金絲桃苷在0.044~0.880μg,異槲皮苷在0.078~1.560μg,槲皮素在0.040~0.800μg,山柰素在0.035~0.700μg線性關(guān)系良好,r均大于0.999 8,平均回收率分別為97.59%、98.97%、100.55%、99.96%。結(jié)論 HPLC指紋圖譜結(jié)合多成分測定可為柿葉及其相關(guān)產(chǎn)品的質(zhì)量控制提供參考。
