近紅外光譜技術(shù)的花生產(chǎn)毒霉菌侵染快速檢測

【摘要】:為了能夠快速、無損地評價花生的質(zhì)量,確保儲藏與食用安全,開發(fā)了一種基于近紅外光譜技術(shù)的花生產(chǎn)毒霉菌污染程度的定性定量分析方法。首先對經(jīng)過Co-60強輻射殺菌后的新鮮花生樣品分別接種谷物中五種常見產(chǎn)毒霉菌(黃曲霉3.17、黃曲霉3.3950、寄生曲霉3.395、寄生曲霉3.0124、赭曲霉3.6486),并于適宜條件下(26℃、RH 80%)儲藏9d。其次,利用近紅外光譜儀采集了不同時期花生樣品在12 000~4 000cm-1波段范圍內(nèi)的漫反射光譜,運用主成分分析(PCA)、判別分析(DA)和偏最小二乘回歸(PLSR)建立了分析模型。結(jié)果顯示,接種不同霉菌的樣品隨著儲藏時間的延長均能得到有效區(qū)分,DA模型對儲藏0,3,6與9d花生的感染單一霉菌和多種霉菌的總體判別正確率分別達(dá)到100%和99.17%,PLSR模型對樣品中的菌落總數(shù)的預(yù)測結(jié)果為:有效決定系數(shù)(R2P)為0.874 1、交互驗證均方根誤差(RMSECV)為0.276Log CFU·g-1,剩余預(yù)測偏差(RPD)為1.92。結(jié)果表明,近紅外光譜技術(shù)可以作為一種可靠的分析方法對花生受霉菌侵染的狀況進(jìn)行快速分析,從而確保貯藏期間花生的質(zhì)量安全。
