pH響應(yīng)型蔗渣木質(zhì)素水凝膠的制備及其對蛋白質(zhì)的控釋性能

【摘要】:該研究以蔗渣木質(zhì)素和甲基丙烯酸為原料合成了pH敏感型蔗渣木質(zhì)素/聚甲基丙烯酸水凝膠,對其合成條件、pH敏感性、溶脹-退溶脹性能以及對牛血清蛋白的控釋等性質(zhì)進(jìn)行研究,并采用紅外光譜、掃描電鏡等對凝膠進(jìn)行表征。結(jié)果表明:(1)對凝膠溶脹比影響的因素由大到小依次為甲基丙烯酸用量、交聯(lián)劑用量、催化劑用量、反應(yīng)的溫度、木質(zhì)素用量。當(dāng)甲基丙烯酸單體濃度為1.75 mol·L~(-1)、木質(zhì)素濃度為25 g·L~(-1)、交聯(lián)劑濃度為3.25×10~(-2)mol·L~(-1)、引發(fā)劑濃度為1.25×10~(-2)mol·L~(-1)、反應(yīng)溫度為65℃時,所得水凝膠在模擬腸液中的溶脹比最大(28.16 g·g~(-1))。與不加木質(zhì)素的聚甲基丙烯酸水凝膠相比,蔗渣木質(zhì)素/聚甲基丙烯酸水凝膠的溶脹比有所下降,但其敏感pH由4~5堿移至6~8。(2)蔗渣木質(zhì)素/聚甲基丙烯酸水凝膠的溶脹—退溶脹可逆性受組成的影響較大,但相對于聚甲基丙烯酸水凝膠,蔗渣木質(zhì)素/聚甲基丙烯酸水凝膠對pH值的敏感響應(yīng)性更強、響應(yīng)速率更快,同時能在更短時間內(nèi)達(dá)到溶脹平衡。(3)加入木質(zhì)素可以提高水凝膠對牛血清蛋白的負(fù)載量,所試驗的蔗渣木質(zhì)素/聚甲基丙烯酸水凝膠樣品對牛血清蛋白的最大負(fù)載量可達(dá)577 mg·g~(-1)。(4)牛血清蛋白在12 h后基本可達(dá)釋放平衡;在模擬胃液中,牛血清蛋白的釋放率僅10%,而在模擬腸液中釋放率達(dá)92%。pH響應(yīng)型蔗渣木質(zhì)素/聚甲基丙烯酸水凝膠可以作為口服型蛋白類藥物的潛在載體。
[Abstract]:The pH sensitive bagasse lignin / polymethacrylic acid hydrogel was synthesized from bagasse lignin and methacrylic acid. The swelling and deswelling properties and the controlled release of bovine serum protein were studied. The gel was characterized by IR and SEM. The results showed that: (1) the factors influencing the swelling ratio of the gel were the amount of methacrylic acid, the amount of crosslinking agent, the amount of catalyst, the reaction temperature and the amount of lignin.
