膠原分子酰化修飾對其結(jié)構(gòu)與性能的影響

【摘要】:在堿性條件下,采用琥珀酸酐(SA)、馬來酸酐(MA)、鄰苯二甲酸酐(PA)對提取的酶溶性膠原進(jìn)行分子修飾,并通過總碳含量、等電點(diǎn)、紅外、X射線衍射、圓二色譜、超靈敏差示掃描量熱以及體外成纖等方法對;昂竽z原的結(jié)構(gòu)與性能進(jìn)行了表征。結(jié)果表明,酸酐改性后的膠原側(cè)鏈羧基含量明顯增加,等電點(diǎn)顯著下降,且;蟮哪z原仍保有特殊的三股螺旋構(gòu)象;不同修飾程度的琥珀酰化膠原的紅外、圓二色譜表明,膠原的酰化程度越高,其三股螺旋構(gòu)象的完整性越差。與PA改性膠原相比,SA、MA;z原有著更完整的三股螺旋構(gòu)象和更高的熱變性溫度。體外成纖實(shí)驗(yàn)顯示,;z原不具備生理?xiàng)l件下的成纖能力,表明三股螺旋結(jié)構(gòu)是膠原成纖的必要不充分條件。
[Abstract]:The enzyme soluble collagen was modified with succinic anhydride, maleic anhydride and phthalic anhydride PAA under alkaline conditions, and was modified by total carbon content, isoelectric point, infrared X-ray diffraction, circular dichroism. The structure and properties of collagen before and after acylation were characterized by hypersensitive differential scanning calorimetry and fiber formation in vitro. The results showed that the carboxyl content of collagen modified by acetic anhydride was obviously increased, the isoelectric point was significantly decreased, and the acylated collagen still retained special three-strand helix conformation. The infrared spectra and circular dichroism of succinylated collagen with different degree of modification showed that the content of carboxyl group in collagen modified by acetic anhydride was obviously increased, and the isoelectric point was decreased significantly. The higher the acylation degree of collagen, the worse the integrity of three helix conformation. Compared with PA modified collagen, the acylated collagen has a more complete triple helix conformation and higher thermal denaturation temperature. In vitro fibrillation experiments showed that acylated collagen did not possess the ability to form fibers under physiological conditions, which indicated that the three-stranded helical structure was the necessary and insufficient condition for the formation of collagen fibers.
