【摘要】:由于氟原子能顯著地改變母體分子的酸堿性、親脂性、構(gòu)象和代謝穩(wěn)定性,各種含氟基團廣泛的出現(xiàn)在藥物分子中。隨著各種氟化試劑的發(fā)展和許多高效的氟化、多氟化反應(yīng)的發(fā)現(xiàn),含氟化合物被應(yīng)用于醫(yī)藥、農(nóng)藥、材料領(lǐng)域。本文主要是對銅催化三氟甲基化和三氟乙基化反應(yīng)的研究。第一部分:銅催化陳氏試劑的三氟甲基化反應(yīng)。我們利用穩(wěn)定,廉價的陳氏試劑為三氟甲基化試劑,成功發(fā)展了一種新的芳香鹵代物的三氟甲基化反應(yīng)。該反應(yīng)只需催化量的無水氯化銅催化,以良好的收率得到了一系列三氟甲基化的產(chǎn)物。該反應(yīng)操作簡單、反應(yīng)時間短、官能團兼容性較好,為陳氏試劑的開發(fā)利用提供了一條新的路徑。第二部分:以亞磷酸二酯為底物,在銅參與下,以氟氯烷烴HCFC-123為三氟乙基源研究了三氟乙基亞磷酸酯合成放大工藝。為消減或淘汰對臭氧層造成破壞的氟氯烷烴HCFC-123做出了一定的貢獻。
[Abstract]:Due to the significant changes in the acidity and alkalinity, lipophilicity, conformation and metabolic stability of the parent molecule, fluorine-containing groups are widely found in the drug molecules. With the development of various fluorinated reagents and the discovery of many efficient fluorination and polyfluorination reactions, fluorinated compounds have been used in medicine, pesticides and materials. In this paper, copper catalyzed trifluoromethylation and trifluoroethylation were studied. Part I: copper catalyzed trifluoromethylation of Chen's reagent. A new trifluoromethylation reaction of aromatic halides was successfully developed by using stable and cheap Chen's reagent as trifluoromethylation reagent. A series of trifluoromethylation products were obtained by using anhydrous copper chloride as catalyst in good yield. The reaction process is simple, the reaction time is short, and the functional group compatibility is good, which provides a new way for the development and utilization of Chen's reagent. The second part: the synthesis and amplification process of trifluoroethyl phosphite was studied with the participation of copper and hydrochlorofluoroalkane hydrocarbon HCFC-123 as trifluoroethyl source. In order to reduce or phase out the ozone layer causing damage to hydrochlorinated hydrocarbon HCFC-123 has made a certain contribution.
