基于菲并咪唑、香豆素和氧雜蒽的新型熒光探針的合成與性能研究

【摘要】:熒光探針具有選擇性好、靈敏度高、響應(yīng)時(shí)間短等優(yōu)點(diǎn),作為一種優(yōu)越的生物分子傳感器,已被廣泛地應(yīng)用于生物成像、疾病診斷、分子識(shí)別及發(fā)光材料等領(lǐng)域。至今為止,基于不同熒光團(tuán)(氧雜蒽、香豆素、氟硼二吡咯等)的用于檢測(cè)生物相關(guān)的陽離子、陰離子和有機(jī)小分子的探針已經(jīng)被廣泛的報(bào)道。本文主要設(shè)計(jì)并合成了兩例基于菲并咪唑的熒光探針(L_1-L_2),兩例基于香豆素的熒光探針(L_3-L_4),一例基于氧雜蒽的熒光探針(L_5)。5個(gè)目標(biāo)化合物(L_1-L_5)均經(jīng)過氫譜(1H NMR)、碳譜(13C NMR)、高分辨質(zhì)譜(HR-MS)的確證結(jié)構(gòu);衔風(fēng)_1和L_2是基于菲并咪唑的識(shí)別HSO_3~-探針,L_1能夠DMSO與水體系中選擇性識(shí)別HSO_3~-,L_2能夠乙醇與水體系中選擇性識(shí)別HSO_3~-;化合物L(fēng)_3和L_4是基于香豆素的HSO_3~-探針,能夠在DMSO和水體系中選擇性識(shí)別HSO_3~-;化合物L(fēng)_5是基于氧雜蒽識(shí)別Cys和GSH的探針,能夠在水體系中選擇性識(shí)別生物硫醇;化合物(L_1、L_5)均成功的應(yīng)用于MCF-7細(xì)胞內(nèi)熒光成像檢測(cè),表明該化合物在生物體系內(nèi)具有潛在的應(yīng)用價(jià)值。
[Abstract]:The fluorescence probe has the advantages of good selectivity, high sensitivity and short response time. As a kind of superior biomolecular sensor, it has been widely used in the fields of biological imaging, disease diagnosis, molecular recognition and luminescent materials. So far, the detection of biological phase based on different fluoranthracene (coumarin, coumarin, flub two pyrrole) The probes of cations, anions and small organic molecules have been widely reported. In this paper, two fluorescent probes based on phenanthrene imidazole (L_1-L_2), two fluorescent probes based on coumarin (L_3-L_4), a fluorescent probe based on oxanthracene (L_5).5 target compounds (1H NMR), carbon (1H NMR), carbon 13C NMR, confirmed structure of high resolution mass spectrometry (HR-MS). Compounds L_1 and L_2 are based on phenanthrene imidazole identification HSO_3~- probes, L_1 can selectively identify HSO_3~- in DMSO and water system, L_2 can selectively recognize HSO_3~- in ethanol and water system, compound L_3 and derivatives are based on coumarin probes, and can be used in the water system and in the water system. Selective identification of HSO_3~-; compound L_5 is a probe based on the identification of Cys and GSH by oxanthracene, which can selectively identify biothiol in the body of water. Compounds (L_1, L_5) have been successfully applied to the fluorescence imaging detection in MCF-7 cells, indicating that the compound has potential application value in the biological system.
