質譜法在藥物立體異構體區(qū)分中的應用及發(fā)展

【摘要】:質譜法廣泛應用于化學、生命科學、藥學等領域,能夠提供分子質量信息,有助于未知物結構的解析,具有分辨率高、信息量大、樣品用量少、靈敏快速等優(yōu)點,在測定有機化合物精確分子質量、結構解析、反應機理等方面發(fā)揮著重要的作用。手性藥物是藥學領域的一個研究熱點,近年來質譜法逐漸被用于藥物手性雜質區(qū)分中,而且隨著質譜技術的不斷發(fā)展和改進,其在手性區(qū)分和分析中越來越受關注。本工作主要介紹了質譜法在手性藥物立體異構體雜質分析中的應用及發(fā)展歷程,闡述了各類質譜技術用于手性異構體分析的原理和特點,其中詳細舉例介紹了質譜動力學方法和離子淌度質譜方法在手性雜質分析中的應用。通過對比分析目前已有的各類手性分析方法的優(yōu)缺點,說明利用質譜法實現(xiàn)手性異構體的區(qū)分和定量測定具有明顯的優(yōu)勢。質譜法用于藥物手性異構體的分離具有快速、不需要復雜的樣品前處理和昂貴的手性柱等特點,極大地提高了手性藥物分析的效率和準確度。隨著質譜分析技術的迅速發(fā)展,質譜法將在多手性中心藥物的手性雜質分離和生物大分子分析(如多糖、多肽)等領域發(fā)揮巨大潛能。
[Abstract]:Mass spectrometry is widely used in the fields of chemistry, life science, pharmacy and so on. It can provide molecular mass information and help to analyze the structure of unknown objects. It has the advantages of high resolution, large amount of information, small amount of sample, quick sensitivity, and so on. It plays an important role in the determination of molecular weight, structure resolution and reaction mechanism of organic compounds. Chiral drugs are a hot research topic in pharmaceutical field. In recent years, mass spectrometry has been used to distinguish chiral impurities. With the development and improvement of mass spectrometry technology, chiral separation and analysis have attracted more and more attention. In this paper, the application and development of mass spectrometry in the analysis of chiral drug stereoisomers are introduced, and the principles and characteristics of various mass spectrometry techniques for the analysis of chiral isomers are described. The application of mass spectrometry kinetic method and ion mobility mass spectrometry in chiral impurity analysis is described in detail. By comparing and analyzing the advantages and disadvantages of all kinds of existing chiral analysis methods, it is shown that the use of mass spectrometry to distinguish and determine chiral isomers has obvious advantages. The separation of drug chiral isomers by mass spectrometry has the advantages of fast separation, no complicated sample pretreatment and expensive chiral column, which greatly improves the efficiency and accuracy of chiral drug analysis. With the rapid development of mass spectrometry, mass spectrometry will exert great potential in the field of chiral impurity separation and biomolecules analysis (such as polysaccharides, polypeptides) of multi-chiral center drugs.
