基于氧雜蒽和萘酰亞胺的新型熒光探針的合成及性能研究

【摘要】:氧雜蒽類染料(羅丹明、熒光素),當(dāng)螺環(huán)結(jié)構(gòu)存在時(shí)熒光很弱且溶液是無(wú)色的,但當(dāng)以開環(huán)形式存在時(shí),會(huì)產(chǎn)生很強(qiáng)的熒光,并且溶液顏色會(huì)發(fā)生顯著的變化。萘酰亞胺類染料具備分子結(jié)構(gòu)簡(jiǎn)單、熱穩(wěn)定性好、光穩(wěn)定性強(qiáng)、發(fā)射波長(zhǎng)適中且易于修飾等優(yōu)點(diǎn),因而也是一種非常理想的熒光載體。本文主要設(shè)計(jì)并合成了兩個(gè)基于羅丹明B的熒光探針(L1-L2),兩個(gè)基于熒光素的熒光探針(L3-L4)和一個(gè)基于萘酰亞胺的熒光探針(L5),進(jìn)一步考察了其光譜性能和在細(xì)胞成像方面的應(yīng)用。目標(biāo)化合物(L1-L5)的結(jié)構(gòu)均經(jīng)過(guò)1H NMR、13C NMR、HRMS的確證;衔風(fēng)1和L2是Fe3+探針,能夠在甲醇與水體系中選擇性識(shí)別Fe3+;化合物L(fēng)3是SO32-探針,化合物L(fēng)4是N2H4探針,化合物L(fēng)5是Cu2+探針。化合物(L1-L5)均成功應(yīng)用于活細(xì)胞內(nèi)熒光成像檢測(cè),表明它們?cè)谏矬w系內(nèi)具有潛在的應(yīng)用價(jià)值。
[Abstract]:Oxygen heteranthracene dyes (Rhodamine, fluorescein), when the snail ring structure exists, the fluorescence is very weak and the solution is colorless, but when it exists in the open ring form, it will produce very strong fluorescence, and the color of the solution will change significantly. Naphthalimide dyes have the advantages of simple molecular structure, good thermal stability, strong light stability, moderate emission wavelength and easy modification, so they are also an ideal fluorescent carrier. In this paper, we designed and synthesized two fluorescent probes based on Rhodamine B, two fluorescent probes based on fluorescein, and a fluorescent probe based on naphthalene imide. The spectral properties and applications in cell imaging were further investigated. The structure of the target compound L1-L5 was confirmed by 1H NMR-13C NMR-HRMS. Compounds L1 and L2 are Fe3 probes, which can selectively recognize Fe3 in methanol and water systems. Compound L3 is SO32- probe, compound L4 is N2H4 probe, and compound L5 is Cu2 probe. Compounds L _ (1-L _ 5) have been successfully applied to the detection of fluorescence imaging in living cells, indicating that they have potential application value in biological systems.
