【摘要】：將喹啉和丹磺酰胺兩種熒光基團(tuán)同時(shí)引入配體L1,利用L1與鋅離子自組裝構(gòu)筑三核鋅有機(jī)-金屬大環(huán)化合物H-1(比率熒光探針),實(shí)現(xiàn)了對(duì)生物分子谷胱甘肽(GSH)的有效識(shí)別。利用紫外光譜、熒光光譜、~1H NMR、ESI-MS等表征方法研究了H-1對(duì)生物分子谷胱甘肽(GSH)的光譜識(shí)別作用。紫外滴定光譜表明,當(dāng)向H-1中加入谷胱甘肽分子后,425 nm處的吸收峰強(qiáng)度降低,320 nm處的吸收峰強(qiáng)度增大,等吸收點(diǎn)為355 nm。利用320 nm處的吸光度值模擬計(jì)算平衡常數(shù),lg K為4.03±0.11,說明H-1與GSH形成了1∶1的包合物。熒光光譜分析表明,當(dāng)向H-1中加入GSH后,以340 nm光激發(fā),波長為513 nm處丹磺酰胺的熒光強(qiáng)度下降,并且發(fā)生紅移,而396 nm處喹啉基團(tuán)的熒光強(qiáng)度增大。利用喹啉基團(tuán)與丹磺酰胺基團(tuán)熒光發(fā)射峰強(qiáng)度變化的比值可以精準(zhǔn)檢測(cè)谷胱甘肽分子,檢測(cè)限可達(dá)到2.5×10~(-6)mol·L~(-1)。
[Abstract]:Two fluorescent groups, quinoline and dansulfonamide, were introduced into ligand L1 at the same time, and trinuclear organo-metal macrocyclic compound H-1 (ratio fluorescence probe) was constructed by self-assembly of L1 and zinc ions. The effective recognition of glutathione (GSH) was achieved. The spectral recognition of glutathione glutathione (GSH) by H-1 was studied by UV and fluorescence spectra. The UV titration spectra show that the absorption peak intensity at 425 nm decreases and increases at 320 nm when glutathione molecule is added to H-1, and the isoabsorption point is 355 nm. The equilibrium constant K is 4.03 鹵0.11 by using the absorbance at 320 nm, which indicates that the inclusion compound between H-1 and GSH is 1:1. Fluorescence spectrum analysis showed that when GSH was added to H-1, the fluorescence intensity of dansulfonamide decreased at 513 nm and red shifted, but the fluorescence intensity of quinoline group increased at 396nm. Using the ratio of quinoline group to Dansulfonamide group fluorescence emission peak intensity, glutathione molecule can be detected accurately, and the detection limit can reach 2.5 脳 10~(-6)mol / L ~ (-1) ~ (-1).
