基于近紅外光譜定量分析植物水解蛋白摻假牛奶

【摘要】:植物水解蛋白是一種典型的蛋白質(zhì)含氮摻假物質(zhì),被不法商販作為廉價(jià)的蛋白質(zhì)添加到牛奶中以提高其蛋白質(zhì)含量。本研究自行配置不同濃度的植物水解蛋白摻假牛奶,應(yīng)用近紅外光譜分析儀漫反射模塊直接掃描摻假樣品,同時(shí)使用透射分析模塊掃描三氯乙酸前處理的摻假樣品;應(yīng)用TQ軟件分別建模并比較兩種方法差異。結(jié)果表明:使用一階導(dǎo)數(shù)Norris Derivative濾波預(yù)處理,在8000~5000 cm~(-1),主成分?jǐn)?shù)10,使用PCR構(gòu)建了最優(yōu)植物水解蛋白漫反射定量分析模型,RMSEC、RMSEP和R分別為0.224、0.214和0.98414,平均回收率為93.2479%。使用一階導(dǎo)數(shù)Norris Derivative濾波預(yù)處理,在10000~7000 cm~(-1),主成分?jǐn)?shù)10,使用PCR構(gòu)建了最優(yōu)植物水解蛋白透射定量分析模型,RMSEC、RMSEP和R分別為0.175、0.138和0.99036,平均回收率為98.7351%。經(jīng)過(guò)三氯乙酸處理的透射模型更加精確和穩(wěn)定,可以用于牛奶中植物水解蛋白的檢測(cè),可以為牛奶品質(zhì)控制和快速定量提供參考。
[Abstract]:Plant hydrolysate protein is a typical nitrogen-adulterated substance, which is added to milk by illegal traders as cheap protein to increase its protein content. In this study, different concentrations of plant hydrolysate adulterated milk were prepared, the adulterated samples were scanned directly by diffuse reflection module of near infrared spectrum analyzer, and the pretreated samples of trichloroacetic acid were scanned by transmission analysis module. TQ software was used to model and compare the two methods. The results showed that the optimal quantitative analysis model of plant hydrolysate protein was constructed by using the first derivative Norris Derivative filter at 8000g / 5000cm ~ (-1) cm ~ (-1) and main fraction of 10. The average recovery was 93.2479 and 0.224414, respectively. The optimal quantitative analysis model of plant hydrolysate protein was constructed by PCR. The optimal transmission quantitative analysis model of plant hydrolysate protein was constructed by using the first derivative Norris Derivative filter at 10 000 ~ 7 000 cm ~ (-1) ~ (-1) and the main fraction of 10. The PCR was used to construct the optimal transmission quantitative analysis model of plant hydrolysate protein, which was 0.175 ~ 0.138 and 0.99036, respectively. The average recovery was 98.7351. The transmission model treated with trichloroacetic acid is more accurate and stable. It can be used for the detection of phytohydrolysate in milk and can provide reference for milk quality control and rapid quantification.
