光果甘草葉中性多糖結(jié)構(gòu)表征及抗氧化活性研究

【摘要】:本文以光果甘草葉為原料,提取得到水溶性多糖GC C,采用DEAE-Sepharose Fast Flow柱層析法對GC C進行分離純化,得到一種中性多糖(GC I),兩種酸性多糖(GC II和GC III),選擇DPPH自由基清除能力、氧自由基吸收能力(ORAC)強的中性多糖GC I,對其進行結(jié)構(gòu)特征分析。采用高效凝膠滲透色譜法(GPC)、紅外光譜法(FT-IR)、PMP柱前衍生-高效液相色譜法(HPLC)、甲基化-氣相色譜質(zhì)譜法(GC-MS)測定GC I的分子量、單糖組成以及糖苷鍵連接方式。GC I含有兩個分子量分布不同的多糖,它們的平均分子量分別為980 ku及20 ku;紅外光譜中4000~500 cm-1波數(shù)范圍內(nèi)出現(xiàn)了多糖的特征峰;GC I由5種單糖組成,包括:甘露糖、鼠李糖、葡萄糖、半乳糖和阿拉伯糖;GC I中→2)-Araf-(1→、→6)-Galp-(1→和→6)-Glcp-(1→殘基含量最高。
[Abstract]:In this paper, the water-soluble polysaccharide GC C was extracted from Glycyrrhiza uralensis leaves. GC C was separated and purified by DEAE-Sepharose Fast Flow column chromatography. A neutral polysaccharide, two acidic polysaccharides, GC II and GC II, were obtained. The free radical scavenging ability of DPPH was selected. Oxygen free radical absorbability (ORAC) is a strong neutral polysaccharide GC I, and its structural characteristics are analyzed. The molecular weight of GC I was determined by high performance gel permeation chromatography (HGPC), FT-IRN PMP precolumn derivatization and high performance liquid chromatography (HPLC). The molecular weight of GC I was determined by methylation gas chromatography-mass spectrometry (GC-MS). There are two polysaccharides with different molecular weight distribution, their average molecular weights are 980ku and 20ku. the characteristic peaks of polysaccharides appear in the wave number range of 4 000 ~ 500 cm-1 in infrared spectrum, GCI is composed of 5 monosaccharides. Among them, mannose, rhamnose, glucose, galactose and arabinose were the highest in 2)-Araf-(1, 6)-Galp-(1 and 6)-Glcp-(1 residues.
