臨床血脂干化學(xué)檢測方法的研究與應(yīng)用

【摘要】:隨著臨床檢驗技術(shù)的快速發(fā)展,在要求結(jié)果準(zhǔn)確穩(wěn)定的同時,又要加快檢驗結(jié)果的速度,為臨床科室提供快速準(zhǔn)確的數(shù)據(jù)。在臨床檢驗科室中,干化學(xué)檢驗技術(shù)在生化分析領(lǐng)域與傳統(tǒng)濕生化分析領(lǐng)域相比,干化學(xué)檢驗技術(shù)具有快速、準(zhǔn)確,干片易保存,便于攜帶,操作檢測簡單方便等優(yōu)點。但干化學(xué)檢驗技術(shù)在國外應(yīng)用比較成熟,國內(nèi)起步較晚,現(xiàn)階段主要依賴國外成熟干片。因此研發(fā)出具有自主知識產(chǎn)權(quán)的干式生化檢驗技術(shù)的干片,對推動干式生化分析技術(shù)的國產(chǎn)化具有重要意義。本文研究建立三種用于快速檢測血液中膽固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL-C)含量的干式生化分析方法。按照測試項目篩選出不同干片載體膜材料,制備成干片,用干式生化分析儀檢測干片,并建立了測定血液中TC、TG、HDL-C含量的標(biāo)準(zhǔn)曲線,對建立三種干片的方法學(xué)進行了線性實驗、批間與批內(nèi)重復(fù)性實驗。運用此方法制備的干片顏色梯度明顯、顯色均勻,重復(fù)性和穩(wěn)定性較好,TC、TG、HDL-C的線性范圍分別為2.59-9.04 mmol/L、0.565-3.39 mmol/L、0.48-2.88 mmol/L。將制備好的干片置于冰箱4℃條件下,在30天內(nèi)重復(fù)性、穩(wěn)定性良好;批內(nèi)與批間重復(fù)性變異系數(shù)都在5%以下,符合臨床檢驗的要求,與臨床濕生化結(jié)果對比,結(jié)果重復(fù)性變異系數(shù)小于5%,可應(yīng)用于臨床檢測。
[Abstract]:With the rapid development of clinical test technology, the rapid and accurate data are provided for the clinical department, while the results are faster and more accurate. In the clinical laboratory, dry chemical inspection technology is fast and accurate in the field of biochemical analysis compared with the traditional wet biochemical analysis field. Dry film is easy to be preserved, easy to carry and simple and convenient to operate. However, the dry chemical inspection technology is more mature in foreign countries and is late at home. At present, it mainly depends on foreign mature dry tablets. Therefore, dry chemical test technology with independent intellectual property right is developed. It is of great significance. In this paper, three dry biochemical analysis methods used to quickly detect the content of cholesterol (TC), triglyceride (TG) and high density lipoprotein (HDL-C) in blood were developed. In accordance with the test items, the membrane materials of different carrier films were screened, dried slices were prepared, dry chemical analyzer was used to detect dry slices, and the determination of TC, TG, H in blood was established. The standard curve of DL-C content has been carried out in linear experiments and in batch and batch repeatability experiments for the establishment of three kinds of dry slices. The color gradient of the dry slices made by this method is obvious, the color rendering is uniform, and the repeatability and stability are good. The linear range of TC, TG and HDL-C is 2.59-9.04 mmol/L, 0.565-3.39 mmol/L, 0.48-2.88 mmol/L. will be prepared, respectively. Under the condition of 4 degrees fridge, the good dry pieces were repeatable and stable in 30 days. The repeatability coefficient of variation within and between batch and batch was below 5%, which accorded with the requirements of clinical test, compared with the clinical wet biochemical results, the result of repeatability variation coefficient was less than 5%, which could be applied to clinical examination.
