【摘要】：建立了黃顙魚、團(tuán)頭魴和草魚血漿、肌肉、肝臟、腎臟中雙去甲氧基姜黃素、去甲氧基姜黃素和姜黃素同時測定的超高效液相色譜紫外檢測法(UPLC-UV)。樣品經(jīng)乙腈(含0.01%乙酸)提取,無水硫酸鈉除水,正己烷去脂等樣品處理,在ACQUIT UPLC BEH C18色譜柱上分離,428 nm波長處測定。雙去甲氧基姜黃素、去甲氧基姜黃素和姜黃素在0.01~5.00 mg/L濃度范圍內(nèi)呈良好的線性關(guān)系,相關(guān)系數(shù)(r2)分別為0.998 7,0.999 8和0.999 6。在空白魚組織中進(jìn)行0.025,0.05,0.50,1.00 mg/kg 4個水平的加標(biāo)回收實驗,3種待測組分的平均回收率為64.7%~102.2%,相對標(biāo)準(zhǔn)偏差為0.69%~10.8%。魚組織中3種待測組分的檢出限(LOD)均為0.010 mg/kg,定量下限(LOQ)均為0.025 mg/kg。應(yīng)用該方法研究了姜黃素在團(tuán)頭魴體內(nèi)的藥代動力學(xué)規(guī)律。
[Abstract]:A method for simultaneous determination of dimethoxycurcumin, desmethoxycurcumin and curcumin in plasma, muscle, liver and kidney of Pelteobagrus fulvidraco, Bream bream and grass carp was established.The samples were extracted by acetonitrile (containing 0.01% acetic acid), treated with anhydrous sodium sulfate, dewatered by n-hexane, and separated at 428nm on ACQUIT UPLC BEH C18 column.The linear range of dimethoxy curcumin, desmethoxycurcumin and curcumin was 0. 01 and 5. 00 mg/L, the correlation coefficient was 0. 0.998 and 0.999 6, respectively.The average recovery rate of the three components to be tested was 64.7% and 102.2%, and the relative standard deviation was 0.69 and 10.8, respectively, at the four levels of 0.025 ~ 0.05 ~ 0.50 ~ 1.00 mg/kg.The detection limit of the three components in fish tissues was 0.010 mg / kg and the lower limit of quantification was 0.025 mg / kg.The pharmacokinetics of curcumin in the body of Megalobrama bream was studied by this method.
