新型次氯酸熒光探針的合成與性能研究

【摘要】:次氯酸在日常生活和工業(yè)生產(chǎn)中應用十分廣泛,然而殘留在水體中的次氯酸會引起水體污染,對人體健康存在潛在的危害。生物體內(nèi)的次氯酸具有重要的生理功能,一旦其含量超出正常的生理水平范圍,會引起一系列疾病。對于環(huán)境樣品及生物體內(nèi)次氯酸的檢測已成為一個重要課題,發(fā)展新型的快速、靈敏、高選擇性次氯酸熒光探針具有重要意義和應用價值。本論文充分運用分子內(nèi)電荷轉(zhuǎn)移機制,設(shè)計合成了五例新型次氯酸熒光探針,并分別在環(huán)境樣品和生物體系中對次氯酸進行檢測。以4-羥基萘酰亞胺作為熒光團,硫代氨基甲酸酯作為識別受體,合成了兩例超靈敏次氯酸熒光探針NDMTC和Lyso-NDMTC。探針NDMTC對次氯酸具有高的靈敏度和選擇性,能在較寬的pH范圍內(nèi)快速識別次氯酸,可用于環(huán)境樣品中次氯酸的檢測,實現(xiàn)了對細胞和小鼠體內(nèi)內(nèi)源性次氯酸的雙光子熒光成像,并可以區(qū)分乳腺細胞和乳腺癌細胞。溶酶體靶向探針Lyso-NDMTC實現(xiàn)了對肝癌細胞溶酶體內(nèi)次氯酸的檢測和熒光成像分析。以三氰基呋喃為母體,修飾上對甲;脚鹚犷l哪醇酯合成了一例長波長次氯酸熒光探針TCF-OCl。探針具有良好的選擇性和水溶性,反應前后出現(xiàn)了明顯的顏色變化(由淺黃色到紅色),可以通過吸收光譜法和熒光光譜法快速檢測次氯酸。探針TCF-OCl成功應用于環(huán)境水體中次氯酸的檢測;探針最大發(fā)射波長在610 nm,實現(xiàn)了巨噬細胞內(nèi)次氯酸的靈敏成像分析。以4-氨基萘酰亞胺作為熒光團,氨乙基硫脲為識別受體,合成了一例比率型次氯酸熒光探針AETU-HOCl。探針的發(fā)射峰在533 nm,加入次氯酸之后發(fā)射峰藍移至473 nm。探針擁有高選擇性,在其它高濃度分析物存在時依然穩(wěn)定,可以在較寬的pH范圍內(nèi)快速識別次氯酸。探針可用于環(huán)境樣品中次氯酸的檢測分析,并實現(xiàn)了細胞外源性和內(nèi)源性次氯酸的比率熒光成像分析。以苯硼酸為識別受體合成一例次氯酸熒光探針BTAT。探針具有好的選擇性和水溶性?赏ㄟ^吸收光譜法和熒光光譜法快速檢測次氯酸。并實現(xiàn)了環(huán)境樣品和細胞核內(nèi)次氯酸的檢測分析。綜上,本文合成的五例新型次氯酸熒光探針,不僅為水環(huán)境和生物體系中次氯酸的檢測提供了新的方法,而且還為次氯酸熒光探針以及其他環(huán)境污染物熒光探針的發(fā)展奠定了理論基礎(chǔ)。
[Abstract]:Hypochlorite is widely used in daily life and industrial production. However, hypochlorite residues in water can cause water pollution and have potential harm to human health.Hypochloric acid has important physiological function, once its content exceeds the normal physiological level, it will cause a series of diseases.It has become an important subject for the detection of hypochloric acid in environmental samples and organisms. It is of great significance and application value to develop new fast, sensitive and highly selective hypochloric acid fluorescent probes.In this paper, five novel hypochlorite fluorescent probes were designed and synthesized by using intramolecular charge transfer mechanism, and the hypochloric acid was detected in environmental samples and biological systems.Using 4-hydroxynaphthalimide as fluorescence group and thiocarbamate as recognition receptor, two highly sensitive fluorescent probes of hypochloric acid (NDMTC and Lyso-NDMTC) were synthesized.The probe NDMTC has high sensitivity and selectivity to hypochloric acid and can quickly recognize hypochloric acid in a wide pH range. It can be used to detect hypochloric acid in environmental samples and realize two-photon fluorescence imaging of endogenous hypochloric acid in cells and mice.And can distinguish between breast cells and breast cancer cells.Lysosomal targeted probe Lyso-NDMTC was used to detect hypochloric acid in lysosome of hepatoma cells.A long wavelength fluorescence probe TCF-OCl was synthesized with tricyanofuran as the parent and modified with p-formyl phenoborborate.The probe has good selectivity and water solubility, and there are obvious color changes (from light yellow to red) before and after the reaction. Hypochloric acid can be detected quickly by absorption spectrometry and fluorescence spectrometry.The probe TCF-OCl has been successfully applied to the detection of hypochloric acid in environmental water, and the maximum emission wavelength of the probe is 610nm.The sensitive imaging of hypochloric acid in macrophages has been realized.Using 4-aminonaphthalimide as fluorescence group and aminoethyl thiourea as recognition receptor, a fluorescent probe AETU-HOCl was synthesized.The emission peak of the probe was 533 nm, and the emission peak blue shifted to 473 nm after hypochloric acid was added.The probe has high selectivity and remains stable in the presence of other high concentrations of analytes. It can quickly recognize hypochloric acid in a wide pH range.The probe can be used for the detection and analysis of hypochloric acid in environmental samples and the ratio fluorescence imaging analysis of exogenous and endogenous hypochloric acid in cells has been realized.A fluorescent probe BTAT was synthesized by using phenylboric acid as the recognition receptor.The probe has good selectivity and water solubility.Hypochloric acid can be detected quickly by absorption spectrometry and fluorescence spectrometry.The detection and analysis of hypochloric acid in environmental samples and nuclei were carried out.In summary, five new hypochlorite fluorescent probes synthesized in this paper not only provide a new method for the detection of hypochloric acid in water environment and biological system, but also provide a new method for the detection of hypochloric acid in water environment and biological system.It also lays a theoretical foundation for the development of hypochloric acid fluorescent probes and other environmental pollutants fluorescent probes.
