【摘要】:八月瓜是木質(zhì)藤本半常綠纏繞植物,含有豐富的黃酮、多酚、微量元素以及齊墩果酸和熊果苷等三萜化合物,廣泛用于醫(yī)藥、食品、化妝品等領(lǐng)域。本文以湘西八月瓜為研究原料,分別探索了八月瓜果皮中總?cè)频淖贤饪梢姺止夤庾V分析方法、超聲波水解提取八月瓜果皮總?cè)频墓に嚄l件、大孔樹脂吸附總?cè)频膭?dòng)力學(xué)和熱力學(xué)研究以及響應(yīng)面優(yōu)化大孔樹脂吸附分離總?cè)频姆蛛x工藝。以八月瓜果皮干粉為研究原料,80%乙醇溶液作為提取劑,采用超聲波輔助法提取果皮中的總?cè)?建立一種紫外可見分光光度法測(cè)定提取液中總?cè)频姆治龇椒。用香草?高氯酸作為顯色劑,545 nm處作為測(cè)定波長(zhǎng),進(jìn)行比色條件優(yōu)化和方法學(xué)考察。結(jié)果表明,加入0.20 mL八月瓜果皮供試品溶液,放置水浴鍋加熱,至溶劑揮發(fā)完畢,隨后分別加入0.30 mL現(xiàn)配置的5%香草醛-冰醋酸溶液和0.70mL高氯酸,在70℃條件下保持15 min,顯色效果最佳。八月瓜果皮提取液中總?cè)频钠骄厥章蕿?01.1%,RSD值為1.2%。該方法快速、準(zhǔn)確,重復(fù)性好,且用該方法測(cè)得的果皮總?cè)坪颗c高效液相色譜法測(cè)得結(jié)果吻合。采用超聲波輔助-水解法提取八月瓜果皮中的總?cè)?通過(guò)單因素實(shí)驗(yàn)和響應(yīng)面優(yōu)化試驗(yàn)考察了pH值、溫度、時(shí)間對(duì)果皮總?cè)铺崛÷实挠绊。結(jié)果表明,優(yōu)化提取工藝參數(shù)為:pH值為5.6,提取溫度為71℃,提取時(shí)間103 min,總?cè)频奶崛÷蕿?7.2%。從7種商用大孔樹脂中篩選出XDA-1型大孔樹脂吸附分離八月瓜果皮中的總?cè)?探討了XDA-1型大孔樹脂對(duì)八月瓜果皮總?cè)频奈教匦?根據(jù)Van’t Hoff方程,研究吸附熱力學(xué)參數(shù)。動(dòng)力學(xué)研究表明,XDA-1大孔樹脂對(duì)八月瓜果皮總?cè)频奈椒蠝?zhǔn)二級(jí)動(dòng)力學(xué)方程描述;熱力學(xué)研究表明,XDA-1大孔樹脂對(duì)八月瓜果皮總?cè)频奈椒螰reundlich等溫吸附方程。吸附焓變?chǔ)0,表明XDA-1型大孔樹脂對(duì)八月瓜果皮總?cè)频奈竭^(guò)程為放熱過(guò)程,且|ΔH|43 k J/mol,說(shuō)明吸附過(guò)程為物理吸附過(guò)程;吉布斯自由能ΔG0表明吸附過(guò)程是自發(fā)的;吸附熵變?chǔ)0,表明該吸附是一個(gè)伴有熵值增加的過(guò)程。研究了XDA-1型大孔樹脂分離八月瓜果皮中的總?cè)。以固液比、吸附溫度、吸附時(shí)間為單因素,用響應(yīng)面優(yōu)化法考察了這三個(gè)因素對(duì)總?cè)铺崛÷实挠绊。結(jié)果表明,在固液比為1:8 g/mL,40℃溫度下吸附315 min,總?cè)频牡寐蕿?3.53%,試驗(yàn)結(jié)果與模型預(yù)測(cè)值吻合。
[Abstract]:Melon is a woody vine semi-evergreen twining plant, rich in flavonoids, polyphenols, trace elements, oleanolic acid and arbutin and other triterpenoids, widely used in medicine, food, cosmetics and other fields.In this paper, the ultraviolet visible spectrophotometric analysis method of total triterpenoids in the peel of Euphorbia indica L. was studied, and the technological conditions of ultrasonic hydrolysis and extraction of total triterpenoids from the peel of Euphorbia melons were studied.The kinetics and thermodynamics of macroporous resin adsorption of total triterpenoids and the optimization of the process of macroporous resin adsorption and separation of total triterpenoids were studied.The total triterpenoids were extracted from the pericarp by ultrasonic assisted extraction with 80% ethanol solution as the raw material. A method was established for the determination of total triterpenoids in the extract by ultraviolet visible spectrophotometry.Vanillin perchloric acid was used as chromogenic agent at 545 nm as the determination wavelength to optimize the colorimetric conditions and investigate the methodology.The results showed that 0.20 mL fruit peel solution was added and heated in a water bath pot until the solvent was volatilized.
