【摘要】:以乙二醇?xì)ぞ厶菫樵?乙酸酐為;瘎,通過N-乙;磻(yīng),制得了新型溫敏性高分子乙;叶?xì)ぞ厶?通過核磁共振氫譜(1H NMR)、傅里葉變換紅外光譜(FTIR)及試管倒置法對(duì)乙;叶?xì)ぞ厶堑慕Y(jié)構(gòu)及溫敏性進(jìn)行了表征,通過掃描電子顯微鏡(SEM)和紫外-可見分光光度計(jì)(UV-Vis)對(duì)水凝膠的微觀形貌和體外藥物釋放性能進(jìn)行了研究.結(jié)果表明,隨著反應(yīng)時(shí)間和乙酸酐與乙二醇?xì)ぞ厶前被柋鹊脑黾?產(chǎn)物的乙酰度逐漸增加;乙;叶?xì)ぞ厶侨芤壕哂袩峥赡鏈孛粜匀苣z-凝膠轉(zhuǎn)變行為,可以通過控制乙;叶?xì)ぞ厶堑囊阴6群腿芤簼舛?使溶膠-凝膠轉(zhuǎn)變溫度處于室溫至體溫(25~37℃)之間;乙酰化乙二醇?xì)ぞ厶撬z具有"高度孔隙化且孔隙之間相互連通"的結(jié)構(gòu)特點(diǎn),通過控制乙酰度和溶液濃度,可使其孔徑大小處于1~40μm范圍內(nèi);乙;叶?xì)ぞ厶撬z的乙酰度為89.90%時(shí),質(zhì)量分?jǐn)?shù)為5%~7%的水凝膠對(duì)抗癌藥物吉西他濱具有緩釋作用,載藥凝膠的釋藥時(shí)間可達(dá)3~5 d.乙;叶?xì)ぞ厶怯型谒幬镝尫偶敖M織工程等領(lǐng)域得到廣泛應(yīng)用.
[Abstract]:Using ethylene glycol chitosan as raw material and acetic anhydride as acylation agent, N-acetylation reaction was carried out. A novel thermo-sensitive macromolecule, acetylated ethylene glycol chitosan, was prepared. The structure and temperature sensitivity of acetylated ethylene glycol chitosan were characterized by 1H NMR, FTIR and tube inversion. The micromorphology of hydrogels and drug release properties in vitro were studied by scanning electron microscopy (SEM) and UV-Vis-UV spectrophotometer. The results showed that with the increase of reaction time and the molar ratio of acetic anhydride to ethylene glycol chitosan amino groups, The acetyl degree of the product increased gradually, and the acetylated ethylene glycol chitosan solution had the thermo-reversible sol-gel transition behavior, which could be controlled by controlling the acetyl degree and the solution concentration of acetylated ethylene glycol chitosan.
