瓜馥木揮發(fā)油化學(xué)成分的GC-MS分析

【摘要】:目的:研究超臨界CO_2流體提取瓜馥木揮發(fā)油的條件,并與水蒸氣蒸餾法提取的瓜馥木揮發(fā)油化學(xué)成分進行比較研究。方法:主要探討壓力、溫度、時間、流量等條件對萃取率的影響,確定超臨界CO_2萃取瓜馥木揮發(fā)油的最佳條件;采用氣相色譜-質(zhì)譜(GC-MS)法對超臨界CO_2流體提取和水蒸氣蒸餾提取的揮發(fā)油進行了化學(xué)成分的分析鑒定,用峰面積歸一化法計算了各成分的相對百分含量。結(jié)果:瓜馥木揮發(fā)油的最佳萃取壓力為30 MPa,溫度為55℃,萃取時間4 h,CO_2流量為30 L/h;解析釜Ⅰ壓力為12 MPa,溫度為50℃;解析釜Ⅱ壓力為5 MPa,溫度為40℃。超臨界CO_2提取的揮發(fā)油中,從解析釜Ⅰ提取物中共鑒定出49種化學(xué)成分,占總量的84.30%,主要成分為β-石竹烯(34.03%);從解析釜Ⅱ提取物中共鑒定21種化學(xué)成分,占總量的87.54%,主要成分為肉豆蔻酸(37.07%);從水蒸氣蒸餾法提取的揮發(fā)油中共鑒定出41種化學(xué)成分,占總量的25.09%,其主要成分為烴類。結(jié)論:兩種提取方法得到的瓜馥木揮發(fā)油組分大致相近,但含量有較大差異,超臨界CO_2萃取法更適合提取瓜馥木揮發(fā)油。
[Abstract]:Objective: to study the conditions of extraction of volatile oil by supercritical CO_2 fluid and compare it with the chemical constituents of volatile oil extracted by steam distillation. Methods: the pressure, temperature and time were studied. The influence of the flow rate on the extraction rate and the optimum conditions for the extraction of volatile oil by supercritical CO_2 were determined. The chemical constituents of essential oil extracted from supercritical CO_2 fluid and steam distillation were analyzed and identified by GC-MS. The relative percent content of each component was calculated by the method of peak area normalization. Results: the optimum extraction pressure of volatile oil was 30 MPA, the temperature was 55 鈩,
