濾紙SERS基底的制備及其用于精漿檢測(cè)的研究

【摘要】:表面增強(qiáng)拉曼光譜技術(shù)作為一種常用的分析方法,具有檢測(cè)靈敏度高、測(cè)試速度快和非侵入性等優(yōu)點(diǎn),已經(jīng)在生物醫(yī)學(xué)、環(huán)境、食品安全等領(lǐng)域中獲得了廣泛關(guān)注。光譜測(cè)量時(shí)使用的增強(qiáng)基底既是檢測(cè)物的承載基底,同時(shí)又對(duì)檢測(cè)信號(hào)的增強(qiáng)有著重要作用。本文以基于濾紙制備的表面增強(qiáng)拉曼散射(SERS)基底為研究對(duì)象,通過(guò)對(duì)其增強(qiáng)性能的研究,獲得一種可實(shí)現(xiàn)簡(jiǎn)單快速制備且靈敏度高、均勻性好、穩(wěn)定性強(qiáng)的濾紙SERS基底的方法,并將這種濾紙基底用于精漿SERS光譜的檢測(cè)中。主要的研究工作如下:1.使用浸泡法制備經(jīng)過(guò)不同處理的濾紙SERS基底,并對(duì)其增強(qiáng)性能進(jìn)行研究。首先獲得了經(jīng)過(guò)不同處理(不同NaCl濃度、不同膠體浸泡時(shí)間)制得的濾紙基底的掃描電鏡圖,對(duì)濾紙SERS基底表面銀納米粒子的分布情況進(jìn)行對(duì)比分析,隨后以R6G為探針?lè)肿訉?duì)制得的濾紙基底的靈敏性、均一性和時(shí)間穩(wěn)定性進(jìn)行了研究。結(jié)果表明,NaCl濃度和浸泡膠體時(shí)間都會(huì)對(duì)銀納米粒子在濾紙基底上的分布產(chǎn)生重要影響,經(jīng)過(guò)濃度為20mM的NaCl溶液預(yù)處理,并在濃縮銀溶膠浸泡1-2h后制得的濾紙SERS基底,其增強(qiáng)因子就可達(dá)到2 107,對(duì)R6G溶液的檢出限為10-7M,顯示出很高的靈敏度。此外,通過(guò)對(duì)濾紙基底均一性和穩(wěn)定性的研究表明,與未經(jīng)過(guò)任何預(yù)處理的濾紙制備的SERS基底相比,經(jīng)過(guò)預(yù)處理制備的濾紙基底也可以呈現(xiàn)出比較好的均一性和時(shí)間穩(wěn)定性。2.將制得的濾紙SERS基底用于精漿SERS光譜的檢測(cè),通過(guò)分析精漿SERS光譜特征峰譜峰強(qiáng)度及強(qiáng)度變化的相對(duì)標(biāo)準(zhǔn)差,比較不同處理?xiàng)l件制備的濾紙基底用于實(shí)際樣品檢測(cè)時(shí)的增強(qiáng)性能和測(cè)試效果差異,對(duì)濾紙SERS基底的可靠性和均一性作出評(píng)價(jià)。結(jié)果表明,使用濃度為20 mM的NaCl溶液預(yù)處理并在銀溶膠中浸泡1-2 h后即可獲得測(cè)試效果很好的濾紙基底。本論文的初步研究成果表明:采用浸泡法可以簡(jiǎn)便地制備出可靠的濾紙SERS基底,而將濾紙使用NaCl溶液進(jìn)行預(yù)處理可大大縮短基底的制備時(shí)間,這種靈敏性高、均勻性好、穩(wěn)定性強(qiáng)的濾紙SERS基底將很有希望應(yīng)用于犯罪現(xiàn)場(chǎng)體液痕跡鑒定、環(huán)境污染物檢測(cè)、食品農(nóng)藥殘留等實(shí)際檢測(cè)中。
[Abstract]:As a common analytical method, surface-enhanced Raman spectroscopy has been widely used in biomedicine, environment, and has the advantages of high detection sensitivity, fast testing speed and non-invasive. The enhanced substrate used in spectral measurement is the carrier substrate for the detection. At the same time, it plays an important role in the enhancement of detection signal. In this paper, the surface enhanced Raman scattering (SERS) substrate based on filter paper is taken as the research object. Through the study of its enhancement performance, a simple and rapid preparation with high sensitivity is obtained. The method of SERS substrate with good uniformity and strong stability is used in the detection of SERS spectrum of seminal pulp. The main research work is as follows: 1. The filter paper SERS substrate with different treatment is prepared by immersion method. At first, the scanning electron microscope (SEM) of filter paper substrate prepared by different treatment (different NaCl concentration, different colloid immersion time) was obtained. The distribution of silver nanoparticles on the surface of filter paper SERS substrate was compared and analyzed, and the sensitivity of the prepared filter paper substrate was obtained by using R6G as probe molecule. The homogeneity and time stability were studied. The results showed that the distribution of silver nanoparticles on the filter paper substrate was significantly affected by the concentration of NaCl and the time of soaking the colloid. The silver nanoparticles were pretreated with 20 mm NaCl solution. The enhancement factor of filter paper SERS prepared by immersion of concentrated silver sol for 1-2 hours can reach 2107, and the detection limit of R6G solution is 10-7M. in addition, the uniformity and stability of filter paper substrate are studied. Compared with the SERS substrate prepared without any pretreatment, the filter paper substrate prepared by pretreatment can also show good uniformity and time stability. 2. The prepared filter paper SERS substrate can be used to detect the SERS spectrum of seminal plasma. By analyzing the relative standard deviation of the peak intensity and intensity change of seminal plasma SERS spectrum characteristic peak, the enhancement performance and testing effect of filter paper substrate prepared under different treatment conditions for actual sample detection were compared. The reliability and uniformity of filter paper SERS substrate were evaluated. The filter paper substrate was obtained by pretreatment with 20 mm NaCl solution and immersion in silver sol for 1-2 hours. The preliminary research results of this paper show that the reliable filter paper SERS substrate can be easily prepared by immersion method. The pretreatment of filter paper with NaCl solution can greatly shorten the preparation time of the substrate. The filter paper SERS substrate with high sensitivity, good uniformity and strong stability will hopefully be used in the identification of body fluid traces at the crime scene and the detection of environmental pollutants.
