【摘要】：為了探究蛋白質(zhì)復(fù)合物的結(jié)構(gòu)與相互作用,建立了蛋白質(zhì)的分子對接方法,從2014年起參加蛋白質(zhì)復(fù)合物結(jié)構(gòu)預(yù)測競賽的預(yù)測和打分競賽.該方法首先采用結(jié)構(gòu)模建方法預(yù)測單體蛋白質(zhì)分子的結(jié)構(gòu),通過快速傅里葉變換進(jìn)行全空間構(gòu)象搜索.然后,優(yōu)化蛋白質(zhì)復(fù)合物構(gòu)象,并采用全原子統(tǒng)計勢函數(shù)進(jìn)行評價.總結(jié)第32~37輪CAPRI競賽結(jié)果發(fā)現(xiàn),該方法在T104、T105、T111和T118比賽中挑選出了L_(RMSD)小于2.0×10~(-10)m的近天然結(jié)構(gòu).通過對比其他國際優(yōu)秀課題組的方法,分析預(yù)測和打分比賽中取得的成績和不足之處,并為后續(xù)的研究提出了改進(jìn)方案與建議.
[Abstract]:In order to explore the structure of protein complexes and interactions, established the molecular docking method of protein, from 2014 onwards in the structures of protein complexes predicted race prediction and scoring contest. This method adopts the structure modeling method of structure of single protein molecules predicted by fast Fourier transform of the whole space conformation search. Then, optimization the conformation of protein complexes, and using all atom potential function was evaluated. Statistical summary of the 32~37 CAPRI competition results show that this method in T104, T105, T111 and T118 were selected to match the L_ (RMSD) is less than 2 * 10~ (-10) near the native structure of M. By comparing with other methods of international outstanding research group the obtained analysis prediction and scoring game of the achievements and shortcomings, and for the subsequent research put forward the improvement scheme and suggestion.
