新型三氰基二氫呋喃喹諾酮類熒光探針在藥物分析中的研究

【摘要】:目的通過(guò)合成新型三氰基二氫呋喃(DCDHF-2-V)電子受體,設(shè)計(jì)了(DCDHF-2-V)—喹諾酮類熒光探針。創(chuàng)建了一種檢測(cè)喹諾酮類藥物含量的新方法,并研究了其最優(yōu)的實(shí)驗(yàn)條件,同時(shí)探討了其增敏原理。方法首先設(shè)計(jì)、合成了一種新型電子受體-新型三氰基二氫呋喃,并通過(guò)一系列的方法表征了此物質(zhì)結(jié)構(gòu)。利用熒光分光光度分析法,首次建立了新型三氰基二氫呋喃-喹諾酮類熒光探針,并用于體外喹諾酮類藥物含量的檢測(cè)。以新型三氰基二氫呋喃為電子受體,配合了新型三氰基二氫呋喃-環(huán)丙沙星、新型三氰基二氫呋喃-諾氟沙星、新型三氰基二氫呋喃-氧氟沙星、新型三氰基二氫呋喃-加替沙星多種荷移絡(luò)合物,成立了喹諾酮類藥物含量測(cè)定的新方法。結(jié)果通過(guò)一定的反應(yīng)條件,合成了一種新型電子受體-新型三氰基二氫呋喃,并通過(guò)一系列的化學(xué)分析和表征確定了其結(jié)構(gòu)。在適宜的反應(yīng)pH、比例、時(shí)間和溫度下,新型三氰基二氫呋喃與喹諾酮類藥物能夠通過(guò)荷移反應(yīng)形成穩(wěn)定的絡(luò)合物,并探討了絡(luò)合物形成的機(jī)理。此法用于體外檢測(cè)喹諾酮類藥物的含量,具有高靈敏度、高選擇性、操作簡(jiǎn)便等優(yōu)點(diǎn)。結(jié)論合成了新型電子受體-新型三氰基二氫呋喃,構(gòu)建了新型三氰基二氫呋喃喹諾酮類藥物熒光探針,確立了新型三氰基二氫呋喃與喹諾酮類藥物絡(luò)合最佳條件,開(kāi)辟了新型喹諾酮類藥物熒光探針的新思路,并且此種熒光探針能夠用于體外測(cè)定喹諾酮類藥物的含量,其結(jié)果準(zhǔn)確有效。
[Abstract]:Objective through the synthesis of new three - two hydrogen furan (DCDHF-2-V) electron acceptor, design (DCDHF-2-V) - quinolone fluorescent probe. A new method for creating a detection of quinolone drug content, and to study the optimal experimental conditions, and discusses its sensitivity principle. Firstly the design and synthesis of a novel three - two new electron acceptor hydrogen furan, and characterized by a series of methods of the material structure. Using fluorescence spectrophotometric method was established for the first time, the new three - two hydrogen furan - quinolone fluorescent probe, and detected with in vitro quinolone content. With a new three cyano two hydrogen furan as electron acceptor with the new three, ciprofloxacin two hydrogen furan - cyano, new three cyano two hydrogen furan - norfloxacin, new three cyano two hydrogen furan - floxacin, new three cyano two hydrogen furan - more than gatifloxacin A charge transfer complex, set up a new method for determination of quinolones. The reaction conditions of the synthesis of a novel electron acceptor model three cyano two hydrogen furan, and their structures were determined by chemical analysis and characterization of a series of pH in the reaction. The proportion, suitable, time and temperature a new three - two hydrogen furan and quinolones by charge transfer reaction to form a stable complex, and to explore the mechanism of complex formation. This method was used for the content of in vitro detection of quinolones, with high sensitivity, high selectivity, easy operation etc.. The new electronic receptor model three - two hydrogen furan synthesis conclusion, construction a new three - two hydrogen furan quinolone fluorescent probe, established the optimum conditions of a new type of three - two hydrogen furan complexed with quinolones, opened up a new quinolone A new idea of the drug fluorescent probe, and the fluorescence probe can be used for the determination of quinolones in vitro, and the results are accurate and effective.
