中藥多糖及人血清中糖蛋白N-糖鏈的毛細(xì)管電泳指紋圖譜研究

【摘要】:該文建立了激光誘導(dǎo)熒光毛細(xì)管電泳(CE-LIF)檢測(cè)中藥多糖和人血清糖蛋白中N-糖鏈的分析方法。利用8-氨基芘-1,3,6-三磺酸鈉(APTS)為柱前熒光衍生試劑,結(jié)合CE分析,可在15 min內(nèi)同時(shí)測(cè)定7種中性單糖。該方法應(yīng)用于中藥制劑和藥物植物中單糖組成和含量的測(cè)定,加標(biāo)回收率為88.0%~102%,相對(duì)標(biāo)準(zhǔn)偏差(RSD)小于3.0%。該方法靈敏度高、準(zhǔn)確性好、實(shí)用可靠,適合于監(jiān)控中藥制劑中糖類物質(zhì)的種類和含量。同時(shí),基于毛細(xì)管凝膠電泳方法(CGE)建立了人血清中糖蛋白的N-糖鏈指紋圖譜。利用中性涂層聚乙烯醇(PVA)有效地抑制電滲流,增加分離時(shí)間,實(shí)現(xiàn)了糖鏈異構(gòu)體的有效分離。對(duì)5μL人血清進(jìn)行毛細(xì)管電泳分析,共分離得到17個(gè)組分。這為進(jìn)一步研究復(fù)雜生物體系血清蛋白的糖基化變化提供了可能性。
[Abstract]:A laser induced fluorescence capillary electrophoresis (CE-LIFs) method for the determination of N-carbohydrate in traditional Chinese medicine polysaccharides and human serum glycoproteins was established in this paper. Using 8-aminopyrene (8-aminopyrene) -1- (3) -3-trisulfonate sodium trisulfonate (APTSs) as pre-column fluorescence derivatization reagent, the method was combined with CE analysis. Seven kinds of neutral monosaccharides can be determined simultaneously in 15 min. The method has been applied to the determination of monosaccharide composition and content in traditional Chinese medicine preparations and medicinal plants. The recovery rate of standard addition is 88.0 and the relative standard deviation (RSD) is less than 3.0. The method has high sensitivity and good accuracy. Practical and reliable, suitable for monitoring the kinds and contents of carbohydrates in traditional Chinese medicine preparations. At the same time, Based on the capillary gel electrophoresis (CGE) method, the fingerprint of human serum glycoprotein was established. The neutral coated polyvinyl alcohol (PVA) effectively inhibited electroosmotic flow and increased the separation time. A total of 17 components were isolated from 5 渭 L human serum by capillary electrophoresis, which provided the possibility for further study on the glycosylation of serum proteins in complex biological systems.
