溫敏含糖嵌段共聚物的合成及其膠束的自組裝

【摘要】:采用酶促法合成含半乳糖單體6-O-乙烯基壬二酸-D-吡喃型半乳糖酯(OVZG),采用可逆加成-鏈斷裂轉(zhuǎn)移自由基聚合法(RAFT聚合)將大分子引發(fā)劑二聚乙二醇單甲醚甲基丙烯酸甲酯(DEGMA)和OVZG有效結(jié)合,制備相對分子質(zhì)量(簡稱分子量)分散系數(shù)(PDI)較低的溫敏性含半乳糖嵌段共聚物PDEGMA-b-POVZG。通過核磁共振氫譜(1 H NMR),傅里葉變換紅外光譜(FT-IR),凝膠滲透色譜(GPC)對聚合物結(jié)構(gòu)進(jìn)行了確認(rèn)表征。通過紫外-可見光譜(UV-Vis)研究表明,共聚物的低臨界溶解溫度(LCST)可以通過共聚單體的比例進(jìn)行調(diào)控,當(dāng)PDEGMA與OVZG的物質(zhì)的量之比為1∶63時(shí),含糖共聚物的LCST值為33℃。在37℃生理溫度下,PDEGMA-b-POVZG可以自組裝形成納米膠束,透射電子顯微鏡(TEM)顯示自組裝形成的聚合物膠束是結(jié)構(gòu)均勻、形貌規(guī)整的球形,通過動(dòng)態(tài)光散射(DLS)測得納米微球的粒徑約為167nm,該溫敏性半乳糖嵌段共聚物在藥物載體方面具有潛在的應(yīng)用前景。
[Abstract]:Enzymatic synthesis of 6-O- vinylazelenedioic acid (6-OVZGN) containing galactose monomethyl propene with diethylene glycol monomethyl ether was carried out by reversible addition-chain break transfer radical polymerization (raft polymerization). Methyl ester acid (DEGMA) and OVZG were effectively combined with each other. The temperature sensitive PDEGMA-b-POVZG copolymer containing galactose block copolymer was prepared with low molecular weight dispersion coefficient (PDI). The structure of the polymer was studied by 1H NMR, FTIR, GPC and FT-IR spectra. The effect of PDEGMA-b-POVZG on the structure of the polymer was studied by means of 1H NMR, Fourier transform infrared spectroscopy (FTIR) and gel permeation chromatography (GPC). Confirmed characterization was carried out. UV-Visanalysis by UV-Vis spectra showed that, When the ratio of PDEGMA to OVZG is 1: 63, the LCST value of sugary copolymers is 33 鈩,
