內(nèi)生真菌綠色生態(tài)法合成納米銀的研究

【摘要】:從石斛中分離得到一株內(nèi)生真菌DO-1,該菌株經(jīng)鑒定為禾谷鐮刀菌(Fusarium graminearum),其菌體在去離子水中浸泡后分泌的代謝產(chǎn)物與硝酸銀作用后,能還原銀離子產(chǎn)生具有抗菌作用的納米銀粒子。以440~450 nm范圍內(nèi)納米銀的特征性等離子吸收峰以及產(chǎn)物抑菌活性為指標(biāo),研究光照條件、溫度以及p H條件對(duì)制備納米銀粒子的影響。黑暗條件和適當(dāng)高溫有助于納米銀粒子的生成及其抑菌活性的增強(qiáng)。隨著p H的增加,納米銀特征性吸收峰不斷增強(qiáng),但抑菌活性呈現(xiàn)先增強(qiáng)再減弱的趨勢(shì),p H 7~8范圍內(nèi)得到的納米銀粒子的抑菌活性較高。調(diào)節(jié)F.graminearum DO-1菌體浸泡液的p H為7,與終濃度為1 m M的Ag NO3溶液于80℃、黑暗條件下反應(yīng)3 h,獲得納米銀粒子,透射電鏡分析顯示制備的納米銀粒子為球形,直徑范圍2~50 nm。應(yīng)用該內(nèi)生真菌代謝產(chǎn)物制備具有抑菌功能的納米銀粒子,方法簡(jiǎn)單,條件溫和,復(fù)合綠色生態(tài)的發(fā)展趨勢(shì)。
[Abstract]:An endophytic fungus DO-1 was isolated from Dendrobium. The strain was identified as Fusarium graminearum. The metabolites secreted by Fusarium graminearum in deionized water reacted with silver nitrate. Silver nanoparticles with antimicrobial activity were produced by reducing silver ions. The characteristic plasma absorption peaks of silver nanoparticles in the range of 440 nm and the bacteriostatic activity of the products were used as indexes to study the illumination conditions. The effects of temperature and pH conditions on the preparation of silver nanoparticles were investigated. Dark conditions and appropriate high temperatures contributed to the formation of silver nanoparticles and the enhancement of their bacteriostatic activities. With the increase of pH, the characteristic absorption peaks of silver nanoparticles increased. However, the bacteriostatic activity of silver nanoparticles obtained in the range of pH 7O 8 was higher than that in the range of p H 7O 8. The pH value of F graminearum DO-1 soaking solution was 7, and that of Ag NO3 solution with the final concentration of 1 mm was 80 鈩,
