海南草豆蔻及其主要活性組分、金屬元素含量的譜學(xué)測(cè)定

【摘要】:采用紅外光譜、紫外光譜、熒光光譜及微波消解電感耦合等離子體質(zhì)譜法對(duì)海南白沙和霸王嶺兩個(gè)產(chǎn)地的草豆蔻進(jìn)行了分析。其乙醇提取物的紅外光譜都具有1 051,1 390,2 976及3 300cm~(-1)左右表征黃酮類成分的特征吸收峰,二階紅外導(dǎo)數(shù)譜圖顯示了在2 977.72及2 899.94有相似吸收峰,在1 922.36及1 650.87cm~(-1)有顯著差異吸收峰,可以鑒定區(qū)分海南不同產(chǎn)地的草豆蔻。利用紫外光譜法表征了兩個(gè)不同產(chǎn)地草豆蔻的特征譜圖,并根據(jù)有關(guān)的吸收原理,建立了測(cè)定草豆蔻植物中主要組分山姜素與豆蔻明含量的方法,獲得了良好的分析精密度(0.42%~0.29%)和回收率(84.28%~117.41%)。測(cè)得白沙草豆蔻中山姜素和豆蔻明的含量分別為4.23%和3.83%,而霸王嶺的分別為3.72%和3.34%。熒光測(cè)定結(jié)果表明,不同產(chǎn)地的草豆蔻乙醇提取物具有相似的熒光發(fā)射及激發(fā)光譜譜圖,熒光強(qiáng)度的差別也再次證明白沙草豆蔻乙醇提取物的濃度大于霸王嶺的。利用微波消解電感耦合等離子體質(zhì)譜法分析了草豆蔻中的K,Ca,Mg,Na,Al,Fe,Mn,Cu,Zn等十七種金屬元素及其含量。該研究為定性、定量地鑒定區(qū)別不同產(chǎn)地或不同種植批次的草豆蔻及測(cè)定其中主要活性組分的含量建立了簡(jiǎn)單可行的方法;根據(jù)實(shí)驗(yàn)測(cè)得的金屬元素種類及含量的結(jié)果,可為進(jìn)一步開(kāi)發(fā)利用草豆蔻及提高食品安全意識(shí)提供更好的理論依據(jù)。
[Abstract]:Infrared spectrum, ultraviolet spectrum, Fluorescence spectra and microwave digestion inductively coupled plasma-mass spectrometry (ICP-MS) were used to analyze cardamom from Baisha and Bawangling, Hainan Province. The infrared spectra of ethanol extracts were characterized by about 1 051C 1 390C 2976 and 3 300 cm ~ (-1) cm ~ (-1) of flavonoids. Characteristic absorption peaks of similar components, The second order infrared derivative spectra showed that there were similar absorption peaks at 2977.72 and 2899.94, and significant differential absorption peaks at 1 922.36 and 1 650.87 cm ~ (-1). It is possible to identify and distinguish the cardamom from different habitats in Hainan. The characteristic spectra of two different areas of cardamom were characterized by ultraviolet spectroscopy, and according to the principle of absorption, A method for the determination of curcumin and cardamine in cardamom was established. A good analytical precision of 0.42 and a recovery rate of 84.28 and 117.41 were obtained. The contents of curcumin and cardamine were 4.23% and 3.83, respectively, and those of Bawangling were 3.72% and 3.34, respectively. The ethanol extracts from different producing areas have similar fluorescence emission and excitation spectra. The difference of fluorescence intensity also proves again that the concentration of ethanol extract of Cortex cardamonis is higher than that of Bawangling. 17 kinds of metal elements and their contents in Caommeg cardamom are analyzed by microwave digestion inductively coupled plasma-mass spectrometry (ICP-MS). The study was qualitative, A simple and feasible method was established to quantitatively identify and determine the contents of main active components of cardamom from different producing areas and different planting lots, and to establish a simple and feasible method for the determination of metal elements and their contents according to the experimental results. It can provide a better theoretical basis for the further development and utilization of cardamom and food safety awareness.
