野薔薇中齊墩果酸和熊果酸的測(cè)定、提取及指紋圖譜研究

【摘要】:野薔薇(Rosa Multiflora Thunb)屬于薔薇科薔薇屬植物,又名多花薔薇,是一種重要的藥食兩用的植物資源,含有豐富營養(yǎng)物質(zhì),具有很高的研究價(jià)值。本文采用HPLC法對(duì)野薔薇中齊墩果酸和熊果酸測(cè)定方法、提取及果實(shí)提取物的指紋圖譜等較為系統(tǒng)地研究,并研究了野薔薇總皂苷對(duì)昆明種小鼠的毒性。通過實(shí)驗(yàn)所得到的實(shí)驗(yàn)結(jié)果如下:1)野薔薇中齊墩果酸和熊果酸分析方法的建立:采用HPLC法測(cè)定野薔薇不同部位(根、莖、果實(shí))中齊墩果酸和熊果酸的含量且比較了12批野薔薇果實(shí)中兩者含量。齊墩果酸和熊果酸在20 min內(nèi)較好分離,其質(zhì)量濃度分別為0.02468~0.2468 mg/m L和0.02523~0.2523 mg/mL時(shí)與峰面積線性關(guān)系良好,相關(guān)系數(shù)分別為0.9994和0.9996;平均回收率為分別為99.96%(RSD為0.74%)和100.03%(RSD為0.95%)。齊墩果酸和熊果酸均在果實(shí)中含量最高;庫車縣、阿圖什市的野薔薇果實(shí)中齊墩果酸和熊果酸的含量最多,可作為最適開發(fā)利用的固定來源。2)響應(yīng)面法優(yōu)化野薔薇果實(shí)中齊墩果酸和熊果酸提取工藝研究:采用超聲波提取法提取野薔薇果實(shí)中齊墩果酸和熊果酸,以單因素試驗(yàn)為基礎(chǔ),結(jié)合響應(yīng)面試驗(yàn)優(yōu)化野薔薇果實(shí)中齊墩果酸和熊果酸的提取工藝。齊墩果酸最佳提取工藝條件為:液料比16:1mL/g,提取溫度41℃,提取時(shí)間37min;熊果酸最佳提取條件為:液料比17:1mL/g,提取溫度40℃,提取時(shí)間38min。在此工藝條件下齊墩果酸和熊果酸實(shí)際得率分別為0.3981 mg/g和0.7170mg/g,與模型預(yù)測(cè)結(jié)果(0.4067mg/g和0.7311mg/g)基本相符。響應(yīng)曲面法優(yōu)化超聲波提取野薔薇果實(shí)中齊墩果酸和熊果酸的預(yù)測(cè)準(zhǔn)確,操作簡單,可以為野薔薇果實(shí)中齊墩果酸和熊果酸的開發(fā)利用提供了參考數(shù)據(jù)。3)野薔薇果實(shí)提取物的HPLC指紋圖譜研究:采用高效液相色譜法研究不同產(chǎn)地野薔薇果實(shí)提取物的指紋圖譜。實(shí)驗(yàn)結(jié)果表明野薔薇果實(shí)提取物的指紋圖譜檢出7個(gè)共有峰,但各批藥材相同成分的含量差異較大,其精密度、穩(wěn)定性、重復(fù)性均良好。該方法可為野薔薇果實(shí)的質(zhì)量控制提供參考。4)野薔薇總皂苷的毒性研究:本研究為了評(píng)價(jià)野薔薇總皂苷的毒性,對(duì)野薔薇總皂苷進(jìn)行毒性實(shí)驗(yàn),從實(shí)驗(yàn)結(jié)果得出,該藥總皂苷提取物L(fēng)D505g/kg,屬無毒化合物。
[Abstract]:Rosa Multiflora Thunb (Rosa Multiflora Thunb), also known as Rosa multiflorus, is an important medicinal and edible plant resource, which contains rich nutrients. In this paper, HPLC method was used to determine oleanolic acid and ursolic acid in rose, extraction and fingerprint of fruit extracts were studied systematically. The toxicity of total saponins of Rosa rose to Kunming mice was studied. The experimental results are as follows: 1). Establishment of analytical method for oleanolic acid and ursolic acid in wild rose: HPLC method was used to determine different parts (roots) of wild rose. The contents of oleanolic acid and ursolic acid in stems and fruits were compared and the contents of oleanolic acid and ursolic acid were compared in 12 batches of Rosa rugosa fruits. Oleanolic acid and ursolic acid were better separated within 20 min. The linear relationship between the concentration and peak area is good when the mass concentration is 0.02468 mg/mL and 0.02523 mg/mL, respectively. The correlation coefficients were 0.9994 and 0.9996, respectively. The average recoveries were 99.96 (RSD = 0.74) and 100.03 (RSD = 0.95), respectively. The contents of oleanolic acid and ursolic acid were the highest in the fruits. The content of oleanolic acid and ursolic acid in roserose fruit of Kuqa County, Atushi City was the highest. Optimization of extraction process of oleanolic acid and ursolic acid from roserose fruit by response surface method: extraction of oleanolic acid and ursolic acid from wild rose fruit by ultrasonic extraction. On the basis of single factor test and response surface test, the extraction process of oleanolic acid and ursolic acid from rose fruit was optimized. The optimum extraction conditions of oleanolic acid were as follows: the ratio of liquid to material was 16: 1 mL / g. The extraction temperature was 41 鈩,
