耐力訓練誘導的肥胖大鼠Irisin抑制脂肪組織FTO表達
[Abstract]:Aim: to study the relationship between the changes of Irisin level and the expression of FTO in adipose tissue of obese rats induced by high fat diet induced by endurance training. Methods: male SD rats were randomly divided into control group (group C), high fat diet group (HD), group) combined with high fat diet group (HDE) group) and exercise training group (CE group). The slope was 0 擄, 8.2m/min (equivalent to 53%VO2max), the first exercise was 15 minutes, the second exercise was 30 minutes, the third exercise was 45 minutes, the fourth time was 60 minutes, the total exercise was 8 weeks, 5 times a week. The weight and fat weight of the rats were measured. Blood Irisin content, skeletal muscle FNDC5mRNA and protein expression, adipose tissue FTO and UCP1 gene mRNA and protein expression. Results: high fat diet significantly increased body weight and fat content, decreased blood Irisin level, significantly decreased FNDC5 expression in skeletal muscle, increased FTO expression in adipose tissue and decreased UCP1 expression in adipose tissue. Endurance training significantly decreased the increase of body weight and fat content induced by high fat diet, significantly increased the decrease of blood Irisin induced by high fat diet, and significantly increased the expression of FNDC5 in skeletal muscle induced by high fat diet. The increase of FTO expression in adipose tissue induced by high fat diet was significantly decreased, while the decrease of UCP1 expression induced by high fat diet was significantly increased. The level of Irisin in blood was negatively correlated with FTOmRNA and protein in adipose tissue, FNDC5mRNA and protein in skeletal muscle were negatively correlated with FTOmRNA and protein in adipose tissue in high fat diet and high fat diet combined exercise group. In exercise group, there was no correlation between FNDCmRNA and FTOmRNA in adipose tissue, but there was a moderate negative correlation between FTOmRNA and protein in exercise group. Conclusion: endurance training rats may inhibit the expression of FTO gene in adipose tissue by releasing Irisin, and then enhance the expression of UCP1.
