常氧和低氧運(yùn)動(dòng)對(duì)Ⅱ型糖尿病大鼠骨骼肌葡萄糖轉(zhuǎn)運(yùn)的影響

【摘要】:1目的觀察低氧運(yùn)動(dòng)對(duì)糖尿病大鼠骨骼肌中胰島素信號(hào)傳導(dǎo)途徑中相關(guān)蛋白含量變化,探究低氧運(yùn)動(dòng)對(duì)糖尿病治療作用可能機(jī)制。2方法將60只6~8周齡SD大鼠隨機(jī)挑選10只作為正常對(duì)照組。其余50只大鼠4周高脂膳食飼養(yǎng),一次性腹腔注射鏈脲佐菌素(STZ)誘導(dǎo)糖尿病模型,空腹血糖≥16.7mmol/L為成模標(biāo)準(zhǔn)。將建模成功的糖尿病30只大鼠隨機(jī)分為3組,分別為低氧運(yùn)動(dòng)組、常氧運(yùn)動(dòng)組和糖尿病組,每組各10只。低氧環(huán)境采用低氧帳篷(MAG10)模擬海拔2500m高度,氧濃度15.0%。運(yùn)動(dòng)組3d跑臺(tái)適應(yīng)性訓(xùn)練之后正式開(kāi)始訓(xùn)練,訓(xùn)練4周,每周6天,低氧運(yùn)動(dòng)組和常氧運(yùn)動(dòng)組每天進(jìn)行1h跑臺(tái)訓(xùn)練。最后一次運(yùn)動(dòng)后大鼠禁食12小時(shí),經(jīng)腹腔注射戊巴比妥鈉麻醉之后腹腔主動(dòng)脈取血,解剖大鼠股四頭肌。測(cè)血液中胰島素(Ins),股四頭肌中葡萄糖轉(zhuǎn)運(yùn)蛋白(GLUT4)、磷酸活化腺苷激酶(AMPK)、磷脂酰肌醇-3激酶(PI3K)和蛋白激酶(PKB)含量,所有數(shù)據(jù)用SPSS19.0軟件進(jìn)行單因素方差分析,實(shí)驗(yàn)結(jié)果用平均數(shù)±標(biāo)準(zhǔn)差表示。3結(jié)果(1)與正常對(duì)照組相比,低氧運(yùn)動(dòng)組和常氧運(yùn)動(dòng)組GLUT4含量顯著降低(p0.05)。與糖尿病組相比,常氧運(yùn)動(dòng)組和低氧運(yùn)動(dòng)組GLUT4含量顯著增加(p0.05)。與低氧運(yùn)動(dòng)組相比,常氧運(yùn)動(dòng)組GLUT4含量略低,但沒(méi)有顯著性差異(P0.05)。(2)與正常對(duì)照組相比,低氧運(yùn)動(dòng)組和常氧運(yùn)動(dòng)組PI3K活性非常顯著性降低(P0.01)。與糖尿病組相比,常氧運(yùn)動(dòng)組和低氧運(yùn)動(dòng)組PI3K活性非常顯著性增加(P0.01)。與低氧運(yùn)動(dòng)組相比,常氧運(yùn)動(dòng)組PI3K活性顯著降低(P0.05)。(3)與正常對(duì)照組相比,低氧運(yùn)動(dòng)組和常氧運(yùn)動(dòng)組PKB活性非常顯著性降低(P0.01)。與糖尿病組相比,常氧運(yùn)動(dòng)組和低氧運(yùn)動(dòng)組PKB活性非常顯著性增加(P0.01)。與低氧運(yùn)動(dòng)組相比,常氧運(yùn)動(dòng)組PKB活性顯著降低(P0.05)。(4)與正常對(duì)照組相比,低氧運(yùn)動(dòng)組和常氧運(yùn)動(dòng)組AMPK活性顯著性降低(P0.05)。與糖尿病組相比,常氧運(yùn)動(dòng)組和低氧運(yùn)動(dòng)組AMPK活性非常顯著性增加(P0.01)。與低氧運(yùn)動(dòng)組相比,常氧運(yùn)動(dòng)組AMPK活性顯著降低(P0.05)。(5)與正常對(duì)照組相比,低氧運(yùn)動(dòng)組和常氧運(yùn)動(dòng)組胰島素含量極其顯著性降低(P0.01)。與糖尿病組相比,常氧運(yùn)動(dòng)組和低氧運(yùn)動(dòng)組胰島素含量顯著性增加(P0.05)。與低氧運(yùn)動(dòng)組相比,常氧運(yùn)動(dòng)組胰島素含量非常顯著性降低(P0.01)。4結(jié)論低氧運(yùn)動(dòng)和常氧運(yùn)動(dòng)都能能顯著提高糖尿病大鼠骨骼肌PI3K、PKB、AMPK活性及GLUT4和胰島素含量,但是低氧運(yùn)動(dòng)刺激對(duì)糖尿病大鼠葡萄糖轉(zhuǎn)運(yùn)影響更顯著。
