4周低氧運動對營養(yǎng)肥胖型大鼠骨骼肌自噬相關因子時序性變化影響的研究

【摘要】:1目的:探討在低氧條件下運動對營養(yǎng)肥胖型大鼠腓腸肌自噬相關因子P62、LC3、Beclin-1表達的影響以及低氧運動誘導骨骼肌細胞自噬發(fā)生的可能機制。2方法:采用高脂飲食誘導SD大鼠建立營養(yǎng)肥胖型大鼠模型,隨后選取建模成功大鼠隨機分為:對照組(C)、低氧訓練1周組(E1)、低氧訓練2周組(E2)、低氧訓練3周組(E3)和低氧訓練4周組(E4),共5組,每組10只。低氧暴露模擬13.6%氧濃度(相當于3500m海拔氧含量),動物跑臺進行訓練,速度為20m.min-1,坡度為0,每天運動1h,周末休息2天,分別訓練1周、2周、3周、4周,最后一次訓練結(jié)束24h后將動物處死并采樣,采用RT-PCR技術(shù)檢測模型大鼠腓腸肌P62、LC3、Beclin-1 mRNA以及westent blot技術(shù)檢測P62、LC3、Beclin-1蛋白在實驗干預前后的變化情況。3結(jié)果:E1、E2、E3組與C組相比Beclin-1 mRNA表達量顯著升高(P0.05),E4組略有下降,Beclin-1蛋白表達量在前兩周逐漸下降,與C組相比E2組下降程度顯著(P0.05),后兩周又逐漸增加,E4組比E2組明顯升高(P0.01);E1組P62 mRNA的相對表達量與C對照組相比增加明顯(P0.05),E2組與E1組相比呈持續(xù)升高趨勢(P0.05),E3組P62 mRNA的相對表達量非常顯著低于E2組和E4組(P0.01)。E3組肥胖大鼠P62蛋白相對表達量與E2組相比顯著升高;E2、E3組、E4組LC3 mRNA相對表達量均顯著高于C組(P0.05),5組肥胖大鼠LC3蛋白中LC3II/LC3I比值變化均不明顯。4結(jié)論:(1)低氧運動能顯著增加自噬相關基因的表達,在一定程度上促進自噬的發(fā)生,但是自噬基因的轉(zhuǎn)錄與自噬蛋白質(zhì)的合成具有不同步性。(2)在低氧訓練的前兩周骨骼肌的自噬程度出現(xiàn)增加的趨勢,但是隨低氧運動時間的延長骨骼肌的自噬程度逐漸降低,可能是機體對低氧刺激逐漸適應的結(jié)果。
[Abstract]:1 Objective: To investigate the effect of exercise on the expression of autophagy related factors P62, LC3, Beclin-1 in the gastrocnemius and the possible mechanism of autophagy induced by hypoxia in hypoxic rats, as well as the possible mechanism of hypoxic exercise induced autophagy in skeletal muscle cells: a hyperlipidemic diet induced SD rats to establish a obese rat model, and then a rat model was selected and the rats were selected randomly. The control group (C), hypoxia training 1 weeks group (E1), hypoxia training group (E2), hypoxia training 3 weeks group (E3) and hypoxic training 4 weeks group (E4), a total of 5 groups, 10 rats in each group. The hypoxia exposure simulated 13.6% oxygen concentration (equivalent to 3500m altitude oxygen content), the animals were trained on the platform, the speed was 20m.min-1, the slope was 0, every day Sports 1H, weekend for 2 days rest for 2 days, respectively training. After 1 weeks, 2 weeks, 3 weeks and 4 weeks, the animals were killed and sampled after the last training 24h. RT-PCR technique was used to detect the gastrocnemius muscle P62, LC3, Beclin-1 mRNA and westent blot technology to detect the P62, LC3, Beclin-1 protein before and after the experimental intervention. (P0.05), the E4 group decreased slightly and the expression of Beclin-1 protein decreased gradually in the first two weeks. Compared with the C group, the decrease in E2 group was significantly higher than that in the E2 group (P0.05), and in the latter two weeks, the E4 group increased significantly (P0.01), and the relative expression of P62 mRNA in the E1 group was significantly higher than that in the control group. The relative expression of P62 mRNA was significantly lower than that in group E2 and E4 group (P0.01).E3 group, and the relative expression of P62 protein in obese rats was significantly higher than that of E2 group; E2, E3 and E4 groups were significantly higher than those in the group of E2. The variation in the ratio of the 5 groups of obese rats was not obvious: (1) the increase of hypoxic activity was significantly increased The expression of autophagy related genes promoted autophagy to some extent, but the transcription of autophagy gene was not synchrony with the synthesis of autophagic proteins. (2) the degree of autophagy in skeletal muscles increased in the first two weeks of hypoxia training, but the degree of autophagy gradually decreased with the prolonged hypoxia exercise time. The result of the body's gradual adaptation to hypoxic stimulation.
