【摘要】：觀察有氧運(yùn)動(dòng)對(duì)肥胖SD大鼠白色脂肪棕色化相關(guān)細(xì)胞因子差異表達(dá)的影響,并探討白色脂肪棕色化的分子機(jī)制。方法:將建模成功的肥胖SD大鼠分為運(yùn)動(dòng)組和肥胖組。運(yùn)動(dòng)組大鼠進(jìn)行8周的跑臺(tái)運(yùn)動(dòng),肥胖組和對(duì)照組不給予運(yùn)動(dòng)。取各組大鼠皮下白色脂肪和肩胛下棕色脂肪,采用肉眼觀察和鏡下觀察;用Real time-PCR檢測(cè)各組脂肪組織中UCP-1、PGC-1α、Irisin和PRDM16 mRNA表達(dá)水平;用Western blot檢測(cè)各組脂肪組織中UCP-1蛋白表達(dá)。結(jié)果:1)通過(guò)肉眼觀察到運(yùn)動(dòng)組SD大鼠腹股溝皮下白色脂肪組織中夾雜著數(shù)塊黃豆大淺棕色的脂肪組織,光鏡下類似棕色脂肪細(xì)胞;Real time-PCR檢測(cè)顯示UCP-1、PGC-1α和irisin mRNA的表達(dá)水平明顯高于肥胖組和對(duì)照組皮下白色脂肪細(xì)胞,Western blot顯示UCP-1蛋白表達(dá)水平明顯高于對(duì)照組和肥胖組皮下白色脂肪細(xì)胞。2)運(yùn)動(dòng)組SD大鼠肩胛間的棕色脂肪組織肉眼觀察平均體積變大,彈性好,鏡下血管豐富,檢測(cè)UCP-1、PGC-1α、irisin mRNA表達(dá)量明顯高于肥胖組和對(duì)照組的棕色脂肪組織。3)運(yùn)動(dòng)組SD大鼠的腹腔及腎周的白色脂肪經(jīng)仔細(xì)檢查和取材均未發(fā)現(xiàn)淺棕色改變。結(jié)論:1)有氧運(yùn)動(dòng)可促進(jìn)肥胖SD大鼠白色脂肪細(xì)胞棕色化;2)有氧運(yùn)動(dòng)提高了SD大鼠肩胛間棕色脂肪的功能,促進(jìn)了細(xì)胞分化;3)有氧運(yùn)動(dòng)對(duì)SD大鼠的腹腔及腹膜后的白色脂肪棕色化的作用微弱,不同部位的白色脂肪可能來(lái)源不同,導(dǎo)致細(xì)胞構(gòu)成和細(xì)胞功能也不同。
[Abstract]:To observe the effect of aerobic exercise on the differential expression of cytokines associated with white fat browning in obese SD rats and to explore the molecular mechanism of white fat browning.Methods: obese SD rats were divided into exercise group and obese group.Rats in exercise group were given treadmill exercise for 8 weeks, while obese group and control group were not given exercise.The subcutaneous white fat and subscapular brown fat were taken from the rats in each group, the expression of UCP-1PGC-1 偽 -Irisin and PRDM16 mRNA in adipose tissue was detected by Real time-PCR, and the expression of UCP-1 protein in adipose tissue was detected by Western blot.Results the white fat tissue of groin subcutaneous of SD rats in exercise group was observed by naked eye.The expression levels of UCP-1PGC-1 偽 and irisin mRNA in brown adipocytes were significantly higher than those in obese group and control group. Western blot showed that UCP-1 protein expression level was significantly higher than that in control group and obese group.The average volume of brown adipose tissue between scapula of SD rats was larger with naked eyes.The expression of UCP-1PGC-1 偽 nirisin mRNA was significantly higher than that in obese group and control group. 3) the white fat in abdominal cavity and perirenal of SD rats in exercise group showed no light brown color changes after careful examination and sampling.Conclusion aerobic exercise can promote the browning of white adipocytes in obese SD rats.) aerobic exercise can improve the function of interscapular brown fat in SD rats.The effect of aerobic exercise on white fat browning in abdominal cavity and retroperitoneum of SD rats was weak, and the white fat in different parts might come from different sources, resulting in different cell composition and cell function.
