IRE1-JNK通路在運(yùn)動(dòng)導(dǎo)致骨骼肌細(xì)胞凋亡中的作用

【摘要】:目的:觀察一次大負(fù)荷運(yùn)動(dòng)對(duì)大鼠骨骼肌細(xì)胞凋亡和內(nèi)質(zhì)網(wǎng)應(yīng)激的影響,以及一次大負(fù)荷運(yùn)動(dòng)后骨骼肌細(xì)胞中JNK,IRE1,TRAF2,ASK1蛋白表達(dá)的變化,探索IRE1-JNK通道在內(nèi)質(zhì)網(wǎng)應(yīng)激引起的骨骼肌細(xì)胞凋亡中的作用。方法:將雄性健康8周齡大鼠,隨機(jī)分為安靜對(duì)照組、運(yùn)動(dòng)組。運(yùn)動(dòng)組的運(yùn)動(dòng)方案為持續(xù)性下坡跑,跑臺(tái)坡度為-16°,速度為16m/min,時(shí)間為90min。根據(jù)取材時(shí)間點(diǎn)不同分為運(yùn)動(dòng)后0h組,12h組,24h組,48h組,72h組。通過(guò)檢測(cè)Caspase3的活性來(lái)觀察細(xì)胞凋亡情況;運(yùn)用western blot檢測(cè)GRP78、JNK、IRE1、TRAF2、ASK1的表達(dá),使用Gel-pro軟件來(lái)進(jìn)行相應(yīng)的條帶的灰度值分析,使用單因素方差分析法處理數(shù)據(jù)。結(jié)果:1.大負(fù)荷運(yùn)動(dòng)后比目魚(yú)肌中的Caspase3的活性在12h達(dá)到峰值;與安靜對(duì)照組相比,除了72h組與安靜組無(wú)顯著性差異之外(P0.05),其余時(shí)相的大鼠比目魚(yú)肌Caspase3的活性都顯著比安靜組高(P0.05)。2.大負(fù)荷運(yùn)動(dòng)后比目魚(yú)肌的GRP78的蛋白表達(dá)在12h達(dá)到峰值;與安靜對(duì)照組相比,大負(fù)荷運(yùn)動(dòng)后比目魚(yú)肌中的GRP78蛋白表達(dá)所有時(shí)相的濃度值都顯著比安靜組高(P0.05)。3.大負(fù)荷運(yùn)動(dòng)后比目魚(yú)肌中的IRE1、TRAF2、ASK1、JNK蛋白表達(dá)在12h達(dá)到峰值;與安靜對(duì)照組相比,大負(fù)荷運(yùn)動(dòng)后比目魚(yú)肌中的IRE1、TRAF2、ASK1、JNK蛋白表達(dá)所有時(shí)相的濃度值都顯著比安靜組高(P0.05)。結(jié)論:一次大負(fù)荷運(yùn)動(dòng)可導(dǎo)致骨骼肌細(xì)胞凋亡,可能是由于運(yùn)動(dòng)引起內(nèi)質(zhì)網(wǎng)應(yīng)激,激活I(lǐng)RE1-JNK通路所致。
[Abstract]:Aim: to observe the effects of a heavy load exercise on apoptosis and endoplasmic reticulum stress in rat skeletal muscle cells and the expression of JNKN IRE1TRAF2 + ASK1 protein in skeletal muscle cells after a heavy exercise. To explore the role of IRE1-JNK channel in apoptosis of skeletal muscle cells induced by endoplasmic reticulum stress. Methods: male healthy rats were randomly divided into quiet control group and exercise group. The slope of treadmill was -16 擄, the speed was 16m / min, and the time was 90 min.According to the different time points after exercise, they were divided into two groups: 0 h group, 12 h group, 24 h group, 48 h group, 72 h group. Apoptosis was observed by detecting the activity of Caspase3, and western blot was used to detect the expression of GRP78, JNKIRE1TRAF2, ASK1. The grayscale value of the band was analyzed by Gel-pro software, and the data were processed by single factor variance analysis. The results showed that the activity of Caspase3 in soleus muscle reached its peak at 12 h after heavy exercise, and compared with that in the quiet control group. Except that there was no significant difference between the 72 h group and the quiet group, the activity of Caspase3 in the soleus muscle of the rest rats was significantly higher than that of the rest group. The protein expression of GRP78 in the soleus muscle reached its peak at 12 h after heavy exercise, and was higher than that in the control group. The expression of GRP78 protein in soleus muscle after heavy exercise was significantly higher than that in control group (P 0.05 路3.The expression of IRE1TRAF2TRAF2ASK1 / JNK protein in soleus muscle reached its peak at 12h, compared with that in quiet control group), and the expression of IRE1TRAF2ASK1 / JNK protein in soleus muscle was significantly higher than that in control group (P < 0.05). The concentration of IRE1TRAF2ASK1 / JNK protein in soleus muscle after heavy exercise was significantly higher than that in control group (P 0.05). Conclusion: one exercise at high load can induce apoptosis of skeletal muscle cells, which may be due to endoplasmic reticulum stress induced by exercise. Activation of IRE1-JNK pathway.
