【摘要】:目的:通過(guò)對(duì)優(yōu)秀自由式摔跤運(yùn)動(dòng)員(運(yùn)動(dòng)級(jí)別都為一級(jí))訓(xùn)練周期血清代謝組學(xué)進(jìn)行分析,同時(shí)結(jié)合傳統(tǒng)生化指標(biāo)檢測(cè),系統(tǒng)全面揭示自由式摔跤這一運(yùn)動(dòng)項(xiàng)目的代謝過(guò)程及其密切相關(guān)的代謝化合物,為科學(xué)的訓(xùn)練監(jiān)控及其合理營(yíng)養(yǎng)干預(yù)提供實(shí)驗(yàn)參考依據(jù)。方法:從山西省自由式摔跤隊(duì)選取10名運(yùn)動(dòng)員(男、女各5人),于冬訓(xùn)開始的第一個(gè)訓(xùn)練周,分別在訓(xùn)練的前期、中期和后期采集3次血液樣本,每次均自肘正中靜脈取血液7m1,2m1放入抗凝管中用于檢測(cè)血常規(guī),剩余5m1全血離心后取血清,份用來(lái)檢測(cè)血清肌酸激酶(CK)、血尿素(BU)和睪酮(T),另一份儲(chǔ)存在-20℃直到進(jìn)行核磁共振信號(hào)采集。通過(guò)1H NMR的代謝組學(xué)方法和多元統(tǒng)計(jì)分析尋找運(yùn)動(dòng)員不同訓(xùn)練階段血清中的差異代謝標(biāo)志物,數(shù)據(jù)均使用SPSS 19.0軟件進(jìn)行單因素方差分析。結(jié)果:(1)常規(guī)生化指標(biāo)檢測(cè)結(jié)果:男運(yùn)動(dòng)員血液WBC、RBC、Hb、Hct和MCV變化趨勢(shì)一致,在訓(xùn)練后都出現(xiàn)下降,經(jīng)過(guò)恢復(fù)之后重新升高,除了WBC恢復(fù)后的指標(biāo)最低值略低于正常范圍外,其余指標(biāo)均處在正常的范圍內(nèi)。血清CK、BU在訓(xùn)練后出現(xiàn)顯著升高(P0.05),CK在恢復(fù)后出現(xiàn)顯著下降(P0.05)。T在訓(xùn)練后和恢復(fù)后也出現(xiàn)了先降低后升高的變化趨勢(shì),但其變化并沒(méi)有顯著性;女運(yùn)動(dòng)員RBC、Hb、Hct和MCV變化趨勢(shì)一致,在訓(xùn)練后都出現(xiàn)下降,經(jīng)過(guò)恢復(fù)之后重新升高,而WBC在訓(xùn)練后和恢復(fù)后則出現(xiàn)連續(xù)下降,除了WBC指標(biāo)訓(xùn)練后和恢復(fù)后的指標(biāo)最低值低于正常范圍外,其余血液指標(biāo)均處在正常的范圍內(nèi)。血清CK訓(xùn)練后與安靜值相比,出現(xiàn)顯著升高(P0.05),恢復(fù)后與訓(xùn)練后相比出現(xiàn)顯著下降(P0.05)。T訓(xùn)練后與安靜值相比出現(xiàn)顯著下降(P0.05),在恢復(fù)后略有升高,但無(wú)顯著性變化。BU在三個(gè)訓(xùn)練階段的值先升高后降低,但均沒(méi)有顯著性變化;(2)代謝組學(xué)檢測(cè)結(jié)果:男運(yùn)動(dòng)員血清代謝組信息中有6種差異代謝標(biāo)志物(P0.05),分別是亮氨酸、異亮氨酸、纈氨酸、p-羥基丁酸、乳酸、馬尿酸鹽。其中與安靜時(shí)相比,男運(yùn)動(dòng)員在訓(xùn)練后血清中的亮氨酸、異亮氨酸、纈氨酸、乳酸顯著升高,而p-羥基丁酸、馬尿酸鹽顯著下降,在恢復(fù)后血清中未發(fā)現(xiàn)有顯著差異的代謝標(biāo)志物。與訓(xùn)練后相比,男運(yùn)動(dòng)員在恢復(fù)后亮氨酸、異亮氨酸、纈氨酸、乳酸發(fā)生顯著下降,而β-羥基丁酸、馬尿酸鹽沒(méi)有顯著性變化;女運(yùn)動(dòng)員血清代謝組信息中有3種差異代謝標(biāo)志物(P0.05),分別是纈氨酸、丙酮酸、肌酸。其中與安靜時(shí)相比,女運(yùn)動(dòng)員在訓(xùn)練后血清中纈氨酸顯著下降,丙酮酸顯著升高,肌酸無(wú)顯著性變化,在恢復(fù)后血清中未發(fā)現(xiàn)有顯著差異的代謝標(biāo)志物。與訓(xùn)練后相比,女運(yùn)動(dòng)員在恢復(fù)后血清中纈氨酸和肌酸含量顯著升高,丙酮酸含量顯著下降。結(jié)論:代謝組學(xué)指標(biāo)同血液常規(guī)生化指標(biāo)均能對(duì)自由式摔跤運(yùn)動(dòng)員的訓(xùn)練監(jiān)控提供有意義的信息,但是采用代謝組學(xué)指標(biāo)相比于血液常規(guī)生化指標(biāo),更能準(zhǔn)確、全面、深刻地反映運(yùn)動(dòng)員訓(xùn)練周期內(nèi)機(jī)體的代謝過(guò)程和規(guī)律,對(duì)指導(dǎo)教練員和運(yùn)動(dòng)員科學(xué)訓(xùn)練和監(jiān)控,進(jìn)行合理營(yíng)養(yǎng)補(bǔ)充具有重要意義。
[Abstract]:Objective: through to the elite free style wrestling (sports level for a training period) serum metabonomics analysis, combined with the detection of traditional biochemical indicators system fully reveal the metabolic process of wrestling the sport and metabolism of closely related compounds, provide experimental reference for the monitoring of the training and reasonable nutrition science the intervention. Methods: a total of 10 athletes from Shanxi province free wrestling team (male and female 5), in the beginning of winter of the first week of training, gather 3 blood samples in the training of the early middle and late respectively, each from the cubital vein blood into 7m1,2m1 anticoagulant tube for blood routine examination. The remaining 5m1 blood serum after centrifugation, samples for detection of serum creatine kinase (CK), blood urea (BU) and testosterone (T), another storage magnetic resonance signal acquisition in -20 until C. By means of 1H NMR metabonomics and multivariate statistical analysis, the serum metabolic markers in different training stages were found. Data were analyzed by one-way ANOVA using SPSS 19 software. Results: (1) the detection results of routine biochemical indexes: WBC, RBC, male athletes of blood Hb, Hct and MCV the same trend, there are decreased after training through recovery after re increased, in addition to the WBC after the recovery of the lowest index slightly lower than the normal range, other indexes were in normal range. The serum CK and BU were significantly increased after training (P0.05), and the CK decreased significantly after the recovery (P0.05). T after training and recovery also emerged after the trend first decreased and then increased, but the change was not significant; female athletes, Hb, Hct RBC and MCV the same change trend, there are decreased after training through recovery after re increased, while WBC after training and recovery appeared after a continuous decline in addition, the WBC index after training and recovery after the index is lower than the normal range, the blood indexes were in normal range. After training, the serum CK was significantly increased (P0.05) compared with the quiet value (P0.05) after the training. After T training, there was a significant decrease compared with the quiet value (P0.05), and a slight increase after recovery, but there was no significant change. BU first increased in the three stage of training value decreased, but there was no significant change; (2) findings of metabolic groups: male athletes serum metabolic information in 6 different metabolic markers (P0.05), respectively, leucine, isoleucine and valine, p- hydroxy butyric acid, lactic acid and hippuric acid salt. Which compared with the quiet, male athletes after training in serum leucine, isoleucine, valine, lactate increased significantly, while p- hydroxybutyric acid, hippurate decreased significantly, found no significant differences in metabolic markers in serum after recovery. Compared with male athletes in training, after the restoration of leucine, isoleucine, valine, lactate significantly decreased, while beta hydroxy butyric acid and hippuric acid salt did not change significantly; female athletes serum metabolic information in 3 different metabolic markers (P0.05), respectively, valine, pyruvate, creatine. Compared with the quiet time, the valine in the serum of female athletes decreased significantly after training, pyruvate increased significantly, and creatine did not change significantly. No significant metabolic markers were found in the serum after recovery. Compared with the training, the content of valine and creatine in the serum of female athletes increased significantly, and the content of pyruvic acid decreased significantly. Conclusion: metabonomics indexes with conventional biochemical indexes of blood of free style wrestling training monitoring provides meaningful information, but the metabonomics index compared to the conventional biochemical index of blood, more accurately and comprehensively and profoundly reflect the metabolic process and regularity of athletes in the training cycle of coaches and body. Athletes scientific training and monitoring, reasonable nutritional supplement has important significance.
