【摘要】：采用液相色譜-串聯(lián)質(zhì)譜(LC-MS/MS)建立了毛發(fā)中司坦唑醇的分析方法,并將其應(yīng)用于單次給藥動物實(shí)驗(yàn)中的動物毛樣品分析。將10mg樣品堿水解后加入戊烷提取,然后進(jìn)行LC-MS/MS分析,采用正離子電噴霧電離、多反應(yīng)監(jiān)測模式測定,方法的最低定量限為25pg/mg。剃去豚鼠背部中央的毛,以60mg/kg的劑量于豚鼠腹腔注射司坦唑醇,然后隔天在同一部位剃取其毛,兩周內(nèi)該豚鼠毛中均能檢出司坦唑醇;給藥后豚鼠毛中司坦唑醇的含量在第1周內(nèi)保持穩(wěn)定,在給藥后第10天達(dá)到峰值。所建立的方法毛發(fā)取樣量少,特異性強(qiáng),靈敏度高,適用于毛發(fā)中司坦唑醇的分析。
[Abstract]:A method for the determination of stetazolol in hair was established by liquid chromatography-tandem mass spectrometry (LC-MS / MS). The method was applied to the analysis of animal hair samples in single dose animal experiment.The 10mg sample was extracted with pentane after alkaline hydrolysis, and then analyzed by LC-MS/MS. The method was determined by positive ion electrospray ionization and multi-reaction monitoring mode. The minimum quantitative limit of the method was 25pg / mg.After shaving the hair in the center of the back of the guinea pig, the guinea pig was injected intraperitoneally with stetazolol at the dose of 60mg/kg, and the hair was shaved at the same place the next day. Within two weeks, the hair of the guinea pig could be detected.The content of stetazolol in guinea-pig hair remained stable in the first week and reached its peak on the 10th day after administration.The method is suitable for the analysis of stetazolol in hair because of its low sampling quantity, high specificity and high sensitivity.
