【摘要】：目的建立犬STR復(fù)合擴(kuò)增體系,為犬個(gè)體識(shí)別和親權(quán)鑒定提供一種檢測(cè)方法。方法用自行建立的11個(gè)犬STR基因座復(fù)合擴(kuò)增體系,使用ABI3130XL遺傳分析儀,對(duì)107份藏獒唾液DNA樣本的擴(kuò)增產(chǎn)物進(jìn)行檢測(cè)及統(tǒng)計(jì)學(xué)分析。結(jié)果除1個(gè)基因座雜合度低于60%,其他10個(gè)基因座雜合度均高于70%;11個(gè)基因座PIC值均在0.6以上;11個(gè)基因座中有8個(gè)基因座的個(gè)體識(shí)別率在0.938以上,其余3個(gè)基因座均在0.826以上;父權(quán)排除率除TETRA為0.267,其余在0.401~0.749之間,偶合機(jī)率1.08×10-13。結(jié)論此11個(gè)犬STR基因座在藏獒犬中具有較高的個(gè)體識(shí)別能力,可用于犬類個(gè)體識(shí)別和親權(quán)鑒定。
[Abstract]:Objective to establish a canine STR complex amplification system for individual identification and paternity identification.Methods the amplified products of 107 samples of saliva DNA from Tibetan mastiff were detected and statistically analyzed by using the ABI3130XL genetic analyzer with 11 canine STR loci multiplex amplification system established by ourselves.Results the heterozygosity of the other 10 loci was higher than 70, the PIC value of 11 loci was above 0. 6, the recognition rate of 8 loci of 11 loci was more than 0.938, and the other 3 loci were above 0.826, the heterozygosity of 1 locus was lower than 60%, the heterozygosity of other 10 loci was higher than 70%.Except for the TETRA of 0.267, the rate of paternity exclusion was between 0.401 and 0.749, and the probability of coupling was 1.08 脳 10-13.Conclusion the 11 canine STR loci have high individual recognition ability in Tibetan Mastiff dogs and can be used for individual identification and paternity identification of dogs.
